Obesity type 2 diabetes mellitus(T2DM)is a common chronic disease that seriously endangers the health of humans and pets.Therefore,it is very important for the pathogenesis and prevention of obesity T2DM.Studies have shown that taurine can promote intestinal development,repair intestinal structure and reduce intestinal inflammation.Therefore,in this study,the obese T2DM rat model induced by high-fat diet combined with streptozotocin(STZ)was used to detect the related indexes of intestinal barrier in obese T2DM rats,in order to reveal the mechanism of taurine in preventing obese T2DM by improving the function of intestinal barrier.In this experiment,8-week-old male SD rats were selected as experimental animals.SD rats were randomly divided into three groups:normal control group,obese T2DM group and taurine group.Animals were adaptively fed for one week before formal feeding.The normal control group was fed with standard rat diet and normal drinking water;Obese T2DM group was fed with high-fat diet and intraperitoneal injection of STZ,and drank normal water;Taurine group was fed with high-fat and injected with STZ,and 2%taurine drinking water was added at the same time.The total test was 10 weeks.After the formal test,SD rats were anesthetized and killed,and the required intestinal tissues were taken for the detection of relevant indexes.Paraffin section were made and stained with HE to observe the changes in the tissue morphology of the duodenum,jejunum and colon.The villus height and crypt depth of the duodenum and jejunum were measured and their ratios were calculated.Total RNA was extracted from intestinal tissue,and the m RNA expression of intestinal nutrients glucose,fatty acid,and amino acid transport related vectors was detected by q RT-PCR;The total protein of intestinal tissue was extracted and the expression of intestinal tight junction related protein was detected by WB;The secretion of proinflammatory related factors and secretory immunoglobulin in intestine were detected by ELISA.Results:1.Taurine can improve the intestinal mucosal structure of obese T2DM rats,and significantly up regulate the protein expression of claudin1 in the mechanical barrier,indicating that taurine can improve the functional state of intestinal mechanical barrier of obese T2DM rats by repairing the integrity of intestinal mucosa and increasing the protein expression of tight junction.2.Taurine can significantly up regulate the protein expression of MUC2,the main component of chemical barrier in obese T2DM rats,indicating that taurine can repair the damage of intestinal chemical barrier in obese T2DM rats.3.Taurine can significantly down regulate the proinflammatory factors IL-6,IL-1β,TNF-αand secretory immunoglobulin s Ig A in the intestinal tissue of obese T2DM rats.This indicates that taurine can improve the intestinal immune barrier of obese T2DM rats.4.Taurine can significantly down regulate the m RNA expression levels of intestinal glucose transport related vectors GLUT2 and SGLT1;Significantly down regulated the m RNA expression levels of fatty acid transport related vectors CD36,Fabp1 and SCL27A4;The m RNA expression levels of neutral amino acid transport related vector(ASCT2,B0AT1),acidic amino acid transport related vector(EAAT3)and basic amino acid transport related vector(ATB0,+,y+LAT2)were significantly up-regulated.The results showed that taurine can improve the transport and absorption of intestinal amino acids in obese T2DM rats,reduce the transport of glucose and lipids in obese T2DM rats,and improve the metabolism of nutrients in obese T2DM rats.The above results indicate that taurine can improve the intestinal mechanical barrier,chemical barrier,immune barrier function of obese T2DM rats,promote the intestinal amino acid transport of obese T2DM rats,inhibit the glycolipid transport,and then alleviate the obesity of obese T2DM rats.
