Huai pig is an important local pig species resource in China.It is distributed in the Huaihe River basin and is an important material for breed selection and new breed cultivation.In recent years,important progress has been made in the selection and breeding of Huai pig strains.Among them,D1,DY,and DH populations have been established in meat quality,multifertility,and resilience,and have been demonstrated and applied in pig production.In order to fully understand the genetic structure of the three strains,understand the genetic basis of their performance differentiation,and further develop the breeding work of the strains,this study counts the number of litter of the D1,DY,and DH breeding groups of Huai pig.Slaughtering determination of succulent traits of male pigs eliminated by breeding groups of 3 strains(8 heads each).The rebreeding group took 30 pigs,collected their hair follicle tissue,and extracted DNA samples;Sixteen random primers of 10 bases were randomly selected for RAPD amplification,and then population genetic structure and genetic distance analysis were performed using Popgen1 32 software.The results show that:(1)The water loss,whiteness,and tenderness of the D1 system were significantly lower than those of the DY and DH systems(P<0.05).The protein content,crude fat content and cooked meat rate were higherP<0.05).In addition to the difference in whiteness indicators,there is no significant difference between DY and DH indicators(P<0.05).(2)38 amplified fragments were amplified by 8 random primers,a total of 485 bands,and the number of strip markers was between 3~7,the number of polymorphic markers was between 3~6,and the size range of marker molecular weight was 0.25~2kb;in all polymorphic bands,18 in D1 accounted for 47.37%,13 in DY for 34.21%,and 13 in DH for 34.21%,Shows obvious genetic differences.(3)Genetic distance analysis showed that the genetic distance between D1 and DY was 0.3507,the genetic distance between DY and DH was 0.0523,and the genetic distance between D1 and DH was 0.3821.The genetic distance between DY and DH was close,and they were far away from the genetic distance of D1,respectively.RAPD clustering results are consistent with those based on meat quality indicators.(4)By sequencing the specific fragments and GenBank-BLAST,7 specific fragments were obtained from X,7,13,14 and 18 chromosomes,respectively.
