Ramie(Boehmeria nivea L.Gaud)is a genus of ramie from the ramie family,which is unique natural fiber crop in China.Chinese rose(Rosa hybrid L.)belongs to the genus Rosaceae,which is a perennial woody plant that has important ornamental value.In order to provide a theoretical basis and technical support for the research on cutting and rapid propagation of ramie and Chinese rose hydroponics,we used the ramie variety “Huazhu No.5” and the rose variety “Pallida” as researched materials to conduct hydroponics cutting rooting experiments.Combine with analysis of related growth,physiological and biochemical indicators to analyze the effects of different position of cutting,hormone treatment,and different concentration of potassium permanganate treatment methods on the rooting of ramie and rose cuttings.The results showed that:1.The site of cutting significantly affected on the rooting characteristics of the ramie.The callus formation of the main stem cutting was 1 day earlier than that of the lateral branch cutting,and the adventitious root formation was nearly 1 week later.The callus rate was 67.78%,and rooting rate was 31.1% of the main stem cutting.While the adventitious roots were formed by collateral cutting,the callus rate reached 87.78% and the rooting rate was 60%.Rooting amount,root length and other indicators were significantly higher than the main stem cuttings,and the lateral branch cuttings had a significantly better rooting effect than the main stem cutting.2.The ramie cutting experiment was conducted by hormone treatment.ABT conducting to the ramie cuttings showed that the 50 ppm of ABT had no obvious accelerating effect on cuttings,and the 50 ppm of ABT had basically the same rooting characteristics as the control,and the rooting quantity index had no significant difference.3.The effect of different concentrations of potassium permanganate on the rooting of ramie cuttings was significant.It was found that ramie cuttings with a cutting depth of 0.5cm were best used for rooting with 1‰ potassium permanganate(D1),which could effectively solve the high-level adventitious roots and rots of ramie cuttings problems.Potassium permanganate impregnated 3cm deep at the base of the cutting,and the rooting effect of ramie cuttings treated for 5h was the worst(D2),while the base of the cutting was soaked with 0.1 ‰ potassium permanganate 5cm,and the effect of cutting rooting at 72h(CK)was middle.4.The potassium permanganate treatment had a significant difference in the root weight index of ramie cuttings,and there was no significant difference in stem and leaf weight index.And there were significant differences in total root length,surface area,volume,and forks of the cuttings that wre treated with the CK,D1 and D2 methods,but there was no significant difference in mean diameter and tips of roots.From the perspective of different level of diameters,the cutting roots of CK,D1,and D2 were mainly concentrated in the diameter of 0.0-1.0mm.Second,ramie cuttings of the formation of adventitious roots start from the 5th day,the fastest growth rate from the 5th day to the 10 th day,and the largest amount of rooting on the 15 th day.And ramie growth rate is D1>CK>D2.5.During rooting process of ramie cuttings,it was found that the SPAD values of ramie cuttings showed a trend was ” increase-decrease-increase” and the root activity of was increased.In the rooting process of ramie,the POD,PPO and IAAO enzyme activities in the rooted area cortex were found to be opposite to the POD,PPO and IAAO enzyme activities in the leaves.The changes of PPO and IAAO enzyme activities in the rooting sites were similar,and the POD enzyme activity had the opposite trend.6.Different concentrations of IAA and SA for cutting roots of the Chinese rose had better effects than control.IAA had a better effect than SA for the rooting of cuttings.The 25mg/LIAA+100mg/LSA and 25mg/LIAA+100mg/LSA conducting to rose cuttings got better rooting effect than single hormone.High concentration of SA treatment inhibited the development of adventitious roots in rose cuttings.7.The adventitious roots of the Chinese rose cuttings were arranged in a circular arrangement at the base of the cuttings,and the rooting type belonged to the callus rooting type.The rooting process could be divided into callus induction period(0-10 days),adventitious root formation period(10-20 days),and adventitious root elongation(after 20days).
