Objective: This study used the method of tissue culture to screen out the most suitable medium of tissue culture each stage of three fine germplasms of Curcuma Kwangsiensis S.G.Lee et C.F.Liang,it provided the experimental basis and theoretical basis for the study of the tissue culture of fine germplasm of Curcuma Kwangsiensis S.G.Lee et C.F.Liang;the compositions of volatile oil of medicinal materials obtaining from the tissue culture of three fine germplasms of Curcuma Kwangsiensis S.G.Lee et C.F.Liang(the following was referred to as tissue culture seedlings)and the medicinal materials of the original germplasm tuber breeding(the following was referred to as raw materials)were determined by GC-MS method,compared the differences between the two,studied on the quality of tissue culture seedlings,it provided experimental basis for the study of the volatile oil components of tissue culture seedlings.Method: This study taked three fine germplasms of Curcuma Kwangsiensis S.G.Lee et C.F.Liang as experimental materials,using the method of tissue culture to screen out the most suitable medium of adventitious bud inductionthree stage? secondary culture stage?tufted bud proliferation stage ? seedling stage ? root induction stage;the compositions of volatile oil of tissue culture seedlings and raw materials were determined by GC-MS method,to compare the differences between the two.Results: 1 ? A28(high oil type),its the most suitable medium of adventitious bud inductionthree stage was MS+6-BA1.0mg/L+ sucrose 30g/L,the induction rate was 83.3%;its the most suitable medium of secondary culture stage was MS+6-BA1.0mg/L+ sucrose 30g/L,the proliferation rate was 2.3;its the most suitable medium of tufted bud proliferation stage was MS+ 6-BA3.0mg/L+ sucrose 30g/L;its the most suitable medium of seedling stage was MS+6-BA3.0mg/L+NAA0.4mg/L+ sucrose 30g/L,the proliferation rate was 2.0;its the most suitable medium of root induction stage was 1/2MS+NAA1.0mg/L+ sucrose 30g/L,the average rooting number was 11.3.2?? 3/B3(high oil and high yield type),its the most suitable medium of adventitious bud inductionthree stage was MS+KT1.0mg/L+ sucrose 30g/L,the induction rate was 76.7%;its the most suitable medium of secondary culture stage was MS+6-BA1.0mg/L+ sucrose 30g/L,the proliferation rate was 2.2;its the most suitable medium of tufted bud proliferation stage was MS+6-BA2.0mg/L+NAA0.1mg/L+ sucrose 25g/L;its the most suitable medium of seedling stage was MS+6-BA3.0mg/L+NAA0.4mg/L+sucrose 30g/L,the proliferation rate was 3.1;its the most suitable medium of root induction stage was 1/2MS+NAA0.4mg/L+ sucrose 30g/L,the average rooting number was 7.1 3?C84(high yield type),its the most suitable medium of adventitious bud inductionthree stage was MS+KT1.0mg/L+ sucrose 30g/L,the induction rate was 70.0%;its the most suitable medium of secondary culture stage was MS+-BA4.0mg/L+ sucrose 30g/L,the proliferation rate was 3.2;its the most suitable medium of tufted bud proliferation stage was MS+6-BA3.0mg/L+ sucrose 30g/L;its the most suitable medium of seedling stage was MS+6-BA3.0mg/L+NAA0.6mg/L+sucrose 30g/L,the proliferation rate was 2.7;its the most suitable medium of root induction stage was 1/2MS+NAA0.4mg/L+ sucrose 30g/L,the average rooting number was 8.8.4?The transplanting survival rate of tissue culture seedlings was above 90%,after planting the tissue culture seedlings and the original germplasm rhizome at the same time,compared to the average yield per plant after harvest: the tissue culture seedlings weight was lower than the raw materials about Curcuma Kwangsiensis S.G.Lee et C.F.Liang,but the tissue culture seedlings weight was obviously higher than raw materials about radix curcumae.5?There were 24 chemical components in the volatile oil contents of A28 tissue culture seedlings and A28 raw materials.The content of Curdione was very high in both,it contained 38.66% in raw materials,it contained 42.75% in tissue culture seedlings.6?There were 21 chemical components in the volatile oil contents of ? 3/B3 tissue culture seedlings and ? 3/B3 raw materials.The content of 2-Cyclohexen-1-one was very high in both,it contained 59.60% in raw materials,it contained 42.19% in tissue culture seedlings.7?There were 18 chemical components in the volatile oil contents of C84 tissue culture seedlings and C84 raw materials.The content of 2-Cyclohexen-1-one was very high in both,it contained 61.19% in raw materials,it contained 50.66% in tissue culture seedlings.Conclusion: This study used the method of tissue culture to screen out the most suitable medium of tissue culture each stage of three fine germplasms of Curcuma Kwangsiensis S.G.Lee et C.F.Liang,the transplanting survival rate of tissue culture seedlings of the three germplasms of Curcuma Kwangsiensis S.G.Lee et C.F.Liang was high,after planting the tissue culture seedlings and the original germplasm rhizome at the same time,compared to the average yield per plant after harvest: the tissue culture seedlings weight was lower than the raw materials about Curcuma Kwangsiensis S.G.Lee et C.F.Liang,but the tissue culture seedlings weight was obviously higher than raw materials about radix curcumae;The volatile oil content of tissue culture seedlings and raw materials were determined by GC-MS method,the components of the tissue culture seedlings were the same as that of the essential oil,and the tissue culture seedlings had basically maintained the excellent characters of the raw materials.
