Since the consecutive dredging for many years, wild resources began to reduce the number of Curcuma kwangsiensis S. G. Lee et C. F. Liang. And germplasm began to decline after a long-term asexual reproduction, the yield reduced and the quality debased. The major object of this study is to establish the rapid propagation system of Curcuma kwangsiensis, and provide theory for industrial propagation in future.The experiment with rhizomatous of Curcuma kwangsiensis S. G. Lee et C. F. Liang as explant, carried on adventitious buds induction, multiplication, seedling cultivation, rooting and transplantation, established the Rapid Propagation system of Curcuma kwangsiensis. S. G. Lee et C. F. Liang.The major results were as following:1. Rapid Propagation: The optimizing medium for multiplication was MS+BA3mg/L+ sucrose 3% + agar0.4%; and MS+BA3mg/L+ MET1.0mg/L +sucrose 3%+ agar0.4% was good for seedling cultivation; the optimizing medium of rooting was MS+NAA2.0mg/L+ sucrose 3%+ agar0.4%. Transplant survival rate was the highest when the test-tube piantlets were treated with MET1.0 mg/L in seedling cultivation, reached 100%.2. Agronomy Characters Research: Among the subsequent generations, supplemented with MET1.0mg/L induced the highest height as 89.7cm, and Chlorophyll content was maximal also,as 1.235 mg/g. The enzyme strip of isozyme of peroxide showed that there was no variation within six generations. Compared the yield, the sixth generation, MET1.0mg/L treatment and the third generation were better than the others. The total weight were 268.1g,240.7g and 218.0g per plant respectively, the weight of root tuber were 180.0g, 157.5g and 133.1g per plant respectively, the number of root tuber were 40, 35 and 31 per plant respectively. Their weight of root tuber per plant and number of root tuber per plant were both higher than the control plant.3. Quality Analysis: Compared the polysaccharide in rhizomatou of each treatment, the extraction rate reached 2.15% which was the highest within all the tissue culture treatments when the subsequent generation was supplemented with MET1.0mg / L .Compared the inorganic elements in root tuber of each treatment, the content of element Cu was 7.75μg/g which was the highest when the subsequent generation was supplemented with 100mg/L Choline Chloride (CC) in seedling cultivation. The content of element Fe and Mn were 94.50μg/g and 109.42μg/g respectively, when the subsequent generations were supplemented with 1.0mg/L MET in seedling cultivation. The content of element Zn,Ca and Mg was highest respectively in the root tuber of the control plant, as72.42μg/g,602.33μg/g and 392.00μg/g respectively. In all, six kinds of inorganic elements in root tuber were higher relatively when the subsequent generations were supplemented with 1.0mg/L MET in seedling cultivation.In summary, MS medium supplemented with BA3.0mg/L as the best Medium in multiplication, supplemented with BA3.0mg/L and MET1.0mg/L was good for plantlets seedling cultivation, supplemented with NAA2.0mg/L was suitable for rooting. In addition, characters of subsequent generations were better when the Plantlets were supplemented with 1.0mg/L MET in seedling cultivation. Pass the test, we can establish the rapid propagation system of Curcuma kwangsiensis S. G. Lee et C. F. Liang, and to realize commercialization of plantlets and industrial production.
