POUIFI (Pituitary Transcription Factor 1) acts as a positive regulatory factor of GH,PRL and TSHβgenes, and it plays an important role in the normal development and growth of mammals.Therefore, it is selected as a candidate gene for growth regulation.In this study, RT-PCRwas used to analyse the extraced RNA from hypophysis in Liang-shan semi-fine wool sheep.The CDs sequence of splicingvariant of the POUIFI gene of the sheep was cloned successfully. The result showed that it contained 418bp with a lack of exon3, exon4 and exon5, so it had no conversed domains. In addition, the splicing variant of the POUIFI geneof this sheep had signal peptide. The secondary structure of the protein is dominate by Random coil, and also contained Helix.QRT-PCR technique was also used in order to study the expression of the POUIFI gene in different tisssues with different growth period.Tissues in six periods were selected randomly, which are 15d,60d,105d,160d,195d and 240d. Each period contained three Liang-shan semi-fine wool sheeps.Longissimus dorsi, liver, heartand brain to collected for analysis. The results showed that the POUIFI gene are expression highest in brain, but very low inLongissimus dorsi, liver and heart.Results of the correlation analysis of mRNA expressions in POUIF1,GH, PRL,TSHR showed that POUIFI regulatedGH, PRL,TSHβexpression by regulating the mRNA expression level, it also hada relationship with the secreting of hormone.
