Modern often used in the treatment of hepatitis, chronic cholecystitis, hyperlipidemia, epilepsy, schizophrenia, tumors, etc. Wen YuJin is one of the widespread use of plant resources, but because of the traditional planting mode of WenYuJin and long-range of vegetative propagation, the big seeding demand, the long growing cycle and untimely disease, lead to the result of variety degeneration and quality reduction. With the use of tissue culture technology, we can not only expedite the breeding of large scale to prevent variety degeneration and save germ plasm-resource of WenYuJin. but also economize manpower and material resources. So. It has great significance to build a system of plant tissue culture. This project has build a systematic and impeccable system of plant tissue culture of WenYuJin. and the further study of quality evaluation and comparative study between the tissue culture WenYuJin and the ordinary WenYuJin not only fill the blank but also provide a certain theoretica to be based on for the steady of quality and clinical practice in the future.The aspect of building systematic and impeccable system of plant tissue culture of WenYuJin:This pr--oject used the good strains of seeds for preparation by screening earlier stage, and found burgeoning buds on the rootstock to be the best explant. Disinfection method is soaking for 13 minute with 75%alcohol and 0.1% mercuric chloride after wipe off the skin of rootstock. The suitable medium of induce recover growin-g of bud is MS+6-BA浓度a3, the other suitable medium of induce multiplication of cluster buds is MS+M浓度f1+NAA浓度e3. After considering-he production and the content of the chemical composition compr-ehensively. then using weight grade me-thod to study the suitable transplanting time and picking time of tissue culture WenYuJin for the first time, result display:the suitable transplanting time of tissue culture WenYuJin is the first transplanting, the suita-ble picking time is from the secone harvest time to the fourth harvest time.The aspect of further comparative study:we have do the further study of quality evaluation and comparative study between the tissue culture WenYuJin and the ordinary WenYuJin for the first time.In terms of plant morphology:in the curse of planting, the tissue culture WenYuJin did not Bloom. except for the comparison of inflorescence and colour between the tissue culture WenYuJin and the ordinary Wen YuJin. the other plant morphologies are in substantial agreement, on the part of medicinal materials characteristic:there are little differences, only some on the side of big or small:on the part of characteristic under Microscope. there are no difference between the up and down surface of foliage and the crossing face of radix and powder in the main. In terms of TLC identification.annual tissue culture and tradition WenYuJin have same characteristic of TLC with reference drugs.Biennia tissue culture and tradition WenYuJin'characteristic of TLC are similar,only in Rf=0.57 has tissue culture Wen YuJin a bright blue spot more than tradition.and both of them are of ten quite different from reference drugs.So reasonable growing period and appropriate harvest time are significant to quality control of Wen YuJin.Research further shows that tissue culture and tradition WenYuJin's moisture content and ash content are in accord with provisions of China pharmacopoeia2010 under YuJin.In terms of extractive substance, In order extractive substance from the most to the lest are biennia tradition WenYuJin, biennia tissue culture WenYuJin, annual tissue culture WenYuJin,annual tradition WenYuJin.With respect to active ingredients, tissue culture WenYuJin's are higher than the corresponding tra-dition's which have same grow ing period in all aspects which include germacrone, curdione, curcuman-oids, naphtha. Especially, Annual tissue culture WenYuJin's germacrone, curdione, curcuminoids are 1.55,4.22,1.41 times higher than tradition's; Biennia tissue culture WenYuJin's curdione are 1.4 times higher than tradition's. In terms of WenYuJin''s germacrone and curdione which have different growing period,the biennia's are all higher than the corresponding annual's:biennia tissue cul-ture's are0.5.0.96 times higher than the corresponding annual's; biennia tradition's are 1.46,3.23 times hi-gher than the corresponding annual's, content of volatile oil decreases slightly. The changes of curcumano-ids are not so obvious.For now, there is not report about developing the rapid propagation system and quality evaluation of tissue culture WenYuJin. The implementation of this issue is first, filling the gaps in this field.
