This study is aim to investigate the optimum reaction conditions of Oyster Polysaccharideide and ZnSO4·7H2O and the effect of Zn-OPS on the growth performance and D-xylose absorption for mice,and explore the effect of Zn-OPS on piglets immune stress by LPS serum antioxidant capacity,intestinal immunity,disaccharidease activity and the flora richness of the intestinal contents,and study the impact of the feed with Zn-OPS on animal intestinal absorption,and the immune stress caused by LPS,then find the best adding amount according to the experimental results and data,provide a theoretical basis as a new green feed additives.1.The Zn-OPS was synthesized from Oyster Polysaccharideide and ZnSO4·7H2O sulfate as raw materials.The formation of Zn-OPS was confirmed by Infrared Spectroscopy.The zinc content was 4.12%determined by Acetylene Flame Atomic Absorption Spectrometry.The optimum reaction condition was analyzed single factor experiment and orthogonal experiment:the optimum reaction temperature of Zn-OPS was 70?,pH was 7,the mass ratio of ZnSO4·7H2O to OPS was 1:2.5,and the reaction time was 60 minutes.2.Fifty male KM mice were randomly divided into five groups according to the principle of similarity of body weight.Group A was fed with basal diet,group B was fed with basal diet 2 800 mg/kg OPS+200 mg/kg ZnSO4.Group C,group D and group E were fed with basal diet containing 1 000 mg/kg,3 000 mg/kg,5 000 mg/kg Zn-OPS for 28 days.The results showed that the growth performance of mice of group B,C,D and E were significantly higher than group A(P<0.05),and the growth performance of mice of group C,D and E were significantly higher than group B(P<0.05).The levels of serum D-xylose of group B,C,D and E were significantly higher than group A(P<0.05),and the levels of serum D-xylose of group C,D and E were significantly higher than group B(P<0.05).The results showed that Zn-OPS increase the growth performance and D-xylose absorption function of mice significantly,and the effect was better after mixed Oyster Polysaccharideide and ZnSO4·7H2O.3.Thirty weaned piglets were randomly divided into 5 groups,group A and B was fed with basal diet,and group C,D and E were fed with 1000,3 000 and 5 000 mg/kg of Zn-OPS.After 30 days of incubation,group B,C,D and E were injected intraperitoneally with 100 ?g/kg LPS,and group A was injected with the same amount of saline.Blood samples were collected at 3 hours after injection,and serum antioxidant indexes were measured.The levels of IL-2,IL-10 and TNF-? and the activity of disaccharides were detected by ELISA.Collected intestine contents and analyze the flora richness of intestinal contents.The results showed that:1)the activity of serum T-SOD and GSH-Px increase significantly with the basal diet adding Zn-OPS(P<0.05),and the levels of MDA decreased significantly(P<0.05).2)Compared with group B,the levels of IL-2 and IL-10 in jejunum mucosa of group C,D and E were significantly increased(P<0.05).The levels of TNF-? in jejunum mucosa of group C and group D were significantly decreased(P<0.05).3)The activities of Sucrase,Maltase and Lactase in jejunum of group C,D and E were significantly increased(P<0.05).4)Compared with group A and B,the intestinal contents of group C,D and E were richer and better,and the number of Lactobacillaceae and other beneficial bacteria in intestinal tract was increased.The results showed that the addition of Zn-OPS could increase the antioxidant capacity of piglets and the immune tolerance of piglets,and improve the activity of intestinal enterotoxin,increase the enrichment of intestinal contents and the number of Lactobacillaceae and other beneficial bacteria,and relief the immune stress significantly.Dietary supplementation of 3 000 mg/kg Zn-OPS showed a better effect.
