ObjectiveTo detect the contents of propolis from different origins.To compare their regulation effects of glucose metabolism in diabetic rats,and to detect the lipid metabolism,energy metabolism,oxidative stress,glucose metabolism-related gene expression.To explore the possible mechanism.Methods1.Detect the contents of different propolisWe choosed seven kinds of propolis,include shandong,Beijing,henan,jiangsu,hangzhou,Brazil and xinjiang propolis.Measurement method is respectively:polyphenols,used Folin-Ciocalteus method;Flavonoids,used the aluminum ion chromogenic method;Vitamin C,used the 2,4-dinitrobenzene hydrazine colorimetric method;Amino acids,used the method of ninhydrin after column;Cinnamic acid,the distribution of column chromatography;For artiprin C,used the method of HPLC;Polysaccharide,used spectrophotometric method;Vitamin B1,B2,used fluorescence spectrophotometry;Vitamin E,coumarin,usd liquid chromatography(HPLC);Calcium,iron,zinc,usd atomic spectrophotometry;Selenium,by hydride atomic fluorescence spectrometry.2.Animal experiment1)Normal rats were randomly divided into two groups,propolis group(propol is 0.17 g/kg)and control group(oil).Each group was treated once daily for 30 days.The level of fasting glucose and glucose tolerance was detected by kits.2)DM rats were induced by STZ,and randomly divided into four groups depends on fastingblood-glucose,that are 0.08 g/kg.bw dose propolis group,0.17 g/kg.bw dose group,0.33 g/kg.bw dose group and control group(oil).Each group was treated once daily for 30 days.The level of fasting blood-glucose and impaired glucose tolerance,body weight and food intake,GHb and GSP were detected.3)Diabetes rats were induced by STZ and randomly devided into 5 groups,that is Beijing,xinjiang,Shandong,Brazilian propolis groups and model group.And set a normal group with normal rats.Detect the body weight and food intake.To detect theindicators related to lipid metabolism,energy metabolism and oxidative stress with kits.And use the method of RT-PCR to detect the expression of related genes.Make the pancreas tissue section.Results1 Propolis from different areas of its contents vary widely.the active ingredient content is relatively rich in Beijing propolis,xinjiang propolis,propolis and Brazilian green propolis in shandong,can be used for subsequent comparative study.2 0.17 g/kg dose of normal rats had no effect on blood glucose and glucose tolerance.3 The best dose to regulate the glucose metabolism is 0.08g/kg.bw.4(1)All the propolis can decrease the fasting blood-glucose,GHb,and GSP of DM rats.But not a significant difference between each other(P>0.05).(2)The level of plasma TG,LDL-c,TC and NEFA are significantly higher than the normal(P<0.01),as well as a significant decrease of HDL-c(P<0.01).(3)The activity of LDH and SDH in the livers and muscle of DM rats were significantly decreased as well as the ATPpase(P<0.05).The Beijing and Brazilian propolis groups' LDH activity and Shandong group's SDH activity had increased to normal level.(4)All the diabetic rats had sever oxidative stress.Four kinds of propolis can at a certain extent enhance the body's antioxidant ability.(5)Brazilian propolis can increase the expression of PPARy and its target genes,namely adiponecin and GLUT4.Brazilian,Beijing and Shandong propolis can repair the damage of pancreas in some degrees.ConclusionsPropolis from different origin had different active ingredient contents.Propolis had no influence on normal rats.All the four propolis can regulate the glucose and lipid metabolism in DM rats with different degrees.The mechanism may be related to antioxidant effects,regulation of metabolic activity,expression of relative genes and so on.
