In recent years,the proportion of household pets is increasing with the improvement of people’s living standards.Followed by pet average age increasing and over nutrition,the incidence of abnormal glucose metabolism diseases increases year by year.People pay more and more attention to such diseases,also to the development and research of related drugs,among which Chinese medicine has shown good curative effect on related diseases.The traditional Chinese medicine Shuanghuangling oral liquid is composed of Astragalus,Raw rehmannia,Poria and other drugs.This prescription is selected according to the clinical symptoms of diabetes and traditional Chinese medicine clinical experience showed a certain effect on regulating blood glucose in the initial clinical application.In order to understand the clinical prevalence and causes of diseases related to abnormal blood glucose in dogs,and to explore the effect of Shuanghuangling on blood glucose regulation in vivo and in vitro,this study was conducted the following parts of the experiment to provide a theoretical basis for future clinical application:Test Ⅰ Investigation of canine clinical cases of diseases related to abnormal blood glucoseIn order to understand the prevalence characteristics of canine abnormal blood glucose-related diseases in pet clinics,and to explore its etiology and influencing factors,this trial conducted a retrospective investigation of 3628 clinical cases in five animal hospitals of Nanjing and Shanghai in the past year.A total of 16 cases with abnormal blood glucose as the main manifestation were collected and analyzed.Through the investigation and analysis of the breed,age,body type,gender,sterilization,diet,feeding method and complications,it is found that the incidence of canine diabetes is more common in clinics,and the incidence rate in the investigated cases is 0.441%.Age,gender,and sterilization have a significant effect on the incidence of diabetes in dogs,that is,older dogs are more susceptible to adult dogs,female dogs are more susceptible to male dogs,sterilized dogs are more susceptible to non-sterile dogs,and the sterilized elder female dogs account for 50%of sick dogs.In addition,obesity,feeding methods and breeds are also important factors that affect the incidence of diabetes in dogs.Diabetes complications are common in sick dogs.Cataract and ketoacidosis are the two most common complications.Test Ⅱ Hypoglycemic effect of Shuanghuangling oral liquid in vitroIn order to initially investigate the hypoglycemic effect of the Shuanghuangling oral liquid,this experiment developed an extraction process based on the drug properties of the Shuanghuangling oral liquid,and determined the inhibitory effects of the prescription and its main components at different dilutions on α-amylase and α-glucosidase activities to investigate the effect regulating blood glucose of the prescription and its components,and the relationship with those concentrations.The results showed that the inhibitory rate of 1 g crude drug/mL Shuanghuangling oral liquid extract on the activity of α-amylase was as high as 98.57%,and the inhibition rate remained above 95%after being diluted 2 and 4 times;.The inhibition rate of Astragalus extract on α-amylase activity is 89.16%,and it decreases with the concentration after dilution.Raw rehmannia extract had no inhibitory effect on α-amylase activity.The Astragalus,Raw rehmannia extracts and Shuanghuangling oral liquid all possessed a good inhibitory effect on α-glucosidase activity,and presented a positive correlation with those concentrations.The Shuanghuangling oral liquid was more effective,and the trend of the inhibitory effect declined with the concentration was relatively gentle.The results showed that the Shuanghuangling oral liquid ’s inhibitory effect on the above two enzyme activities was stronger than that of its constituents of Astragalus and Raw rehmannia.It is inferred that the prescription had a certain effect on regulating blood glucose and could be further investigated as a blood glucose regulating drug.Test Ⅲ The regulation of Shuanghuangling oral liquid on canine blood glucoseIn order to further investigate the effect of the Shuanghuangling oral liquid regulating blood glucose,4 healthy Beagle dogs without glucose metabolism disease were taken as subjects,and fed with the Shuanghuangling oral liquid extract solution at a daily dose of 0.5 g per kg body weight for 4 consecutive days,Before and after medication,after fasting for 12 h,each dog was fed with glucose solution at 2 g per kg of body weight.Blood samples were collected at 15 min,30 min,60 min,90 min and 120 min before feeding and after glucose solution was fed to measure blood glucose value,oral glucose tolerance,and serum Ins,GC,IDE,SOD,GSH-PX activity,MDA,TNF-αand other indicators before and after taken the drug.The results showed that the Shuanghuangling oral liquid had no effect on the fasting blood glucose of the dog and the time required for the blood glucose recovery after glucose fed.The peak blood glucose increased after taken the drug,but it did not exceed the normal range.Serum GSH-PX activity,TNF-α concentration increased significantly after fed the drug(p<0.05).IDE concentration increased very significantly when fasting,there was no significant change after feeding sugar,and significantly increased when the blood glucose value returned to fasting level.The results showed that Shuanghuangling oral liquid help to increase blood glucose under fasting conditions and down regulate blood glucose after feeding,and owned a bidirectional regulation effect on blood glucose.
