With the rapid development of China's economy and the ever-changing urban spatial pattern, coupled with the rapid development of high-speed rail, this puts new demands on the function and layout of coach stations in large cities. The theory of quantity, scale and functional layout of coach stations in big cities needs to be updated urgently.Firstly, the paper analyzes the development history of coach stations in big cities in China,as well as the development of coach station layout and passenger flow organization theory in major cities, and summarizes the main factors affecting the functional layout of coach stations in large cities and the new demands of passenger flow organizations. The relationship between the coach station and the urban form and scale layout was determined, and the relationship between the hierarchical structure and functional structure of the coach station and the number of coach stations and the convenience of passenger travel in the new era was determined.Secondly, with the minimum passenger travel cost and minimum travel time as the optimization goal, the optimization model of passenger flow distribution in the metropolitan coach station is constructed, and the optimization scheme of passenger flow organization in the metropolitan coach station is designed. Finally, based on the Nanjing coach station and the passenger flow organization, based on a large number of on-the-spot investigations and extensive data collection, the distribution status and radiation range of Nanjing coach station,as well as the source of passenger flow and passenger's arriving at the coach station, and the matching of Nanjing coach station and passenger flow organization are analyzed. The optimization plan for the functional layout of the Nanjing coach station was proposed.Studies have shown that the number and layout of coach stations in large cities must be adjusted and reformed in the face of traditional transportation organization models, especially for the connection model between passenger transport chains and local public transport. On the other hand, the long-distance transport passenger flow is further diversified into a multi-level transport structure, which needs to be further refined and improved in terms of traffic organization.
