Sanghuangporus is a large perennial fungus with important medicinal value,named after parasitic on mulberry.Polysaccharides and flavonoids are two important active components in Sanghuangporus.It is widely concerned because of its antitumor,antioxidation,immunomodulatory,bacteriostatic and other biological activities.In this paper,the three Sanghuangporus fruit bodies of Changbai Mountain are used as raw materials to study the optimal extraction technology of three Sanghuangporus polysaccharides and flavonoids.In addition,the differences of antioxidant and bacteriostatic abilities of three kinds of Sanghuangporus are studied through antioxidant and bacteriostatic experiments.Finally,a comparative study of three flavonoids from Sanghuangporus by HPLC.The results are as follows:1.Ultrasonic assisted hot water extraction is used to extract Sanghuangporus polysaccharides.The optimum extraction process of three kinds of Sanghuangporus polysaccharides is determined by single factor test and orthogonal analysis.Sanghuangporus vaninii: ratio of material to liquid 1:40,extraction time 5 h,extraction temperature 100 ?.Under this condition,the polysaccharides content is 7.51 mg/g.Phellinus pini: ratio of material to liquid 1:60,extraction time 5 h,extraction temperature 80 ?.Under this condition,the polysaccharides content is 7.40 mg/g.Sanghuangporus baumii: ratio of material to liquid 1:60,extraction time 4 h,extraction temperature 100 ?.Under this condition,the polysaccharides content is 4.86 mg/g.The three Sanghuangporus are S.vaninii,Phellinus pini and S.baumii in order of polysaccharides content.2.Ultrasonic assisted ethanol extraction is used to extract Sanghuangporus flavonoids.The optimum extraction process was determined by single factor test and response surface analysis.S.vaninii: ratio of material to liquid 1:32,ethanol concentration 62 %,ultrasonic temperature 72 ?.Under this condition,the flavonoids content is(11.489±0.153)mg/g.Phellinus pini: ratio of material to liquid 1:33,ethanol concentration 70 %,ultrasonic temperature 64 ?.Under this condition,the flavonoids content is(4.327±0.192)mg/g.S.baumii: ratio of material to liquid 1:70,ethanol concentration 65 %,ultrasonic temperature 73 ?.Under this condition,the flavonoids content is(1.419±0.093)mg/g.The three Sanghuangporus are S.vaninii,Phellinus pini and S.baumii in order of flavonoids content.3.The results of studies on the antioxidant activity of three Sanghuangporus polysaccharides indicate: Under the same concentration,the total reducing power of polysaccharides from S.vaninii is the highest,followed by that of polysaccharide from Phellinus pini,followed by that of polysaccharide from S.baumii.The hydroxy radical scavenging ability of the three Sanghuangporus polysaccharides are as follows: S.vaninii polysaccharides,Phellinus pini polysaccharides,and S.baumii polysaccharides.Studies on the scavenging of superoxide anion radicals by three polysaccharides from Sanghuangporus showed that in the concentration range of 100-200 ?g / mL,the scavenge rate of polysaccharides from S.vaninii is higher than that of Phellinus pini,and that of S.baumii is the lowest.After the concentration is 200?g / mL,the clearance rate is in the order of clove S.baumii polysaccharides > S.vaninii polysaccharides>Phellinus pini polysaccharides.The order of DPPH and ABTS free radical scavenging ability of three kinds of Sanghuangporus polysaccharides are: S.vaninii polysaccharides >S.baumii polysaccharides >Phellinus pini polysaccharides.Studies have shown that S.vaninii polysaccharide has the best antioxidant effect,S.baumii polysaccharide is second,and Phellinus pini polysaccharide is the worst.The results of studies on the antioxidant activity of three Sanghuangporus flavonoids indicate: The total reducing power of the three Sanghuangporus flavonoids is from S.vaninii flavonoids,Phellinus pini flavonoids,and S.baumii flavonoids.The hydroxy radical scavenging ability of the three Sanghuangporus flavonoids are as follows: S.vaninii flavonoids,Phellinus pini flavonoids,and S.baumii flavonoids.The superoxide anion radical scavenging ability of the three Sanghuangporus flavonoids are as follows: S.vaninii flavonoids,S.baumii flavonoids,and Phellinus pini flavonoids.The order of DPPH and ABTS radical scavenging ability of three kinds of Sanghuangporus polysaccharides are: S.vaninii flavonoids>Phellinus pini flavonoids>S.baumii flavonoids.The order of inhibition of xanthine oxidase activity by the three flavonoids is Phellinus pini flavonoids> S.baumii flavonoids> S.vaninii flavonoids.Studies have shown that S.vaninii flavonoids has the best antioxidant effect,Phellinus pini flavonoids is second,and S.baumii flavonoids is the worst.The results of studies on the antibacterial activity of three Sanghuangporus polysaccharides and flavonoids indicate: The three Sanghuangporus polysaccharides and flavonoids have a certain antibacterial effect on Staphylococcus aureus ?Escherichia coli?Staphylococcus epidermidis?Pseudomonas aeruginosa,but have no obvious inhibitory effect on Candida albicans.4.Through HPLC analysis of the three Sanghuangporus flavonoids,the similarities and differences between different separation methods and the composition of different flavonoids are compared.By HPLC analysis,S.vaninii contains Rutin,Eriodictyol,Quercetin,Naringenin,and Apigenin.Phellinus pini contains Rutin,Eriodictyol,Quercetin,Naringenin,Sakuranetin.S.baumii contains Rutin,Eriodictyol,Naringenin,Apigenin.Through similarity analysis,the similarities between the three Sanghuangporus varieties and the control map are 0.545,0.821,and 0.835,respectively.By cluster analysis,Phellinus pini and S.baumii are clustered into one group and separated from S.vaninii.Through the principal component analysis of three Sanghuangporus,several compounds affecting the quality differences of the three Sanghuangporus are identified.The most important ones are the compounds with retention times of 11.291,17.416,38.827 and 65.168.The compound with a retention time of 38.827 is Naringenin.
