Antioxidants have the abilities which could resist various diseases.Since the organisms in nature can't defend environmental aggression,it is necessary to obtain antioxidants from environment in order to enhance the antioxidant level.In this study two species,Sanghuangporus quercicola and S.lonicericola,were used to study the conditions of solid culture and liquid culture.Then,analyzing the in vitro antioxidant activity of fermentation broth the content of antioxidant active substances.The main results are listed as follows.1.Results of solid culture: The best carbon source for the S.quercicola and S.lonicericola was glucose,and the best nitrogen source was yeast soaking powder.The optimum pH value was 6 and 7 respectively.In the suitable conditions,the hyphae grew fast.2.Results of liquid culture: 1)Single factor results: the volume of addition of glucose was 20 g/L and the yeast extract powder was 5 g/L,and when the pH is less than or equal 7,the two species grow rapidly.2)Results of response surface analysis: for S.quercicola,the carbon source was added 21g/L,the mycelial biomass was 6.627 g/L,the nitrogen source was added 5g/L and the pH value was 7.For S.lonicericola,the carbon source was added 21g/L,the nitrogen source was added 5 g/L,the pH value was 7 and the mycelial biomass was 4.880g/L.The analysis results of the two Sanghuangporus were smaller than the actual predicted values,which verified the model.The parameters given were highly fitted to the actual results,which could better reflect the relationship between each factor and the secondary metabolites of hyphae.3.Antioxidation experiment results of fermentation broth: 1)Determination of antioxidant capacity: Hydroxyl radical scavenging rate of S.lonicericola was superior to that of S.quercicola,and superoxide anion removal rate of S.quercicola was significantly greater than that of S.lonicericola.There was no significant difference between the maximum clearance of the two species.However,the scavenging rate of S.quercicola was higher than that of S.lonicericola,the total antioxidant capacity was better than that of the small hole S.lonicericola,the iron ion reduction ability in the late stage of culture was better than that of S.lonicericola,and the chelating ability of the ferrous ion was inferior to that of S.lonicericola,but the maximum value of the two was not significantly difference.2)Antioxidant active substance: Analyzed by experimental results,the content of polysaccharide,flavonoids andascorbic acid was related to superoxide anion,total antioxidant capacity,DPPH free radical scavenging ability,ABTS free radical scavenging ability.The polyphenol content was related to hydroxyl free base,iron ion reduction ability,ferrous ion chelation ability.The superoxide dismutase content played an auxiliary role in antioxidant capacity.The results of this study showed that both S.quercicola and S.lonicericola had a high level of antioxidant activity.We compared the advantages and disadvantages of the antioxidant experiments in detail.This study stated that the Sanghuangporus fungi could be converted into natural antioxidants in the future.
