Ergothioneine is a natural amino acid with a variety of physiological activities and has a stable antioxidant effect in physiological environments.At present,its development and application are mainly focused on functional foods and medicines,and relatively little research on its application in cosmetics,and most of the studies on ergothioneine are crude extracts,and there are few evaluations on the efficacy of ergothioneine products.In this paper,ergothioneine was used as an index to optimize the extraction process of Pleurotus citrinopileatus crude extract.The anti-oxidation,whitening activity,and antioxidant stability of the ergothioneine and crude extract and refined liquid of Pleurotus citrinopileatus.Based on this,a cosmetic supplemented with Pleurotus citrinopileatus extract was prepared,which provided a basis for the research and development of new edible fungus cosmetics.Ergothioneine to extract as an index,and the single factor optimized by response surface method,mushroom extract obtained citrinopileatus optimum extraction conditions: extraction time for 30 min,extraction temperature 80℃,solid to liquid ratio of 1:30,the extract The ergothioneine content is 0.116mg/m L.The extract was purified using ultrafiltration,ultrafiltration optimum conditions: 5KDa ultrafiltration membrane,0.2bar pressure of ultrafiltration 4h,ergothioneine transmittance was 90.26%.The content of ergothione in the lyophilized water extract of the Pleurotus citrinopileatus after purification by ultrafiltration purification and DM130 resin decolorization was 5.85%,and the chroma was reduced by 80%.The ergothioneine has an IC50 value of 3.03 μ g/m L for hydrogen peroxide clearance,an IC50 value of 33.64 μ g/m L for hydroxyl radical clearance,and an IC50 value of 159.35 μg/m L for superoxide anion clearance.Its antioxidant capacity is equivalent to Vc.The IC50 value of the hydrogen peroxide removal rate of refined liquid of Pleurotus citrinopileatus is 3.53 μ g/m L,in which the ergothioneine content is 0.21 μg / m L;the IC50 of the hydroxyl radical removal rate of refined liquid form Pleurotus citrinopileatus is 0.33mg/m L,where the ergothioneine content is 19.6μg/m L;the IC50 value of the superoxide anion clearance of refined liquid of Pleurotus citrinopileatus is 2.52mg/m L,where the ergothioneine content is147.5 μg/m L.Ergothioneine,water extract and refined liquid of Pleurotus citrinopileatus showed stable antioxidant capacity under the influence of light,high temperature,acid-base conditions and metal ions.Through tyrosinase inhibition and B16-F10 cell whitening experiments,it was found that the ergothioneine inhibits the tyrosinase monophenolase activity with L-tyrosine as the substrate,the IC50 value is 0.29 μg/m L;The IC50 value of inhibition of bisphenolase activity using L-dopa as a substrate was 1.33 μg/m L.The refined liquid of Pleurotus citrinopileatus inhibits the tyrosinase monophenolase activity with L-tyrosine as the substrate,the IC50 value was 3.56 μg/m L,and the IC50 value for inhibition of bisphenolase activity using L-dopa as a substrate was 14.59 μg/m L.The whitening experiment of B16-F10 cells showed that the refined liquid of Pleurotus citrinopileatus was non-toxic in the concentration range of 0.25-10 mg / m L.When the solution concentration reached 10 mg / m L(ergothione content was 0.58 mg / m L),the tyrosinase activity inhibition rate reached 27.82% and the melanin inhibition rate reached 43.9%,all of which were better than the ergothioneine whitening ability of 0.75 mg / m L.The refined liquid of Pleurotus citrinopileatus was used in the formula of toner and lotion with the addition amount of 0.35%,and compared with the addition of 0.02% ergothioneine.According to the patch test,sensory evaluation,stability and moisturizing and anti-oxidan ability of the product,it was found that the cosmetics added with 0.35% refined liquid of Pleurotus citrinopileatus did not cause allergic reactions in the human body,the cosmetic’s moisturizing performance was improved,and the physicochemical properties were stable.Compared with the two competing products in the market that are mainly anti-oxidant,the anti-oxidant ability of the toner added with the refined liquid of Pleurotus citrinopileatus is better than that of the competing products.
