Development Of Whitening Cosmetic Of Traditional Chinese Medicine And Research Of Whitening Mechanism Of Siwu Decoction

"Siwu Decoction" is generally used to main treat the inadequate symptom of the blood, which doses can be plus or minus according to the diseases. Traditional Chinese Medicine(TCM)theory holds that Siwu Decoction can remove blood vessels blockage, balance the body, increase blood circulation, and make ruddy complexion. From the perspective of the modern theory of whitening investigated Siwu Decoction whitening effect, explored its whitening mechanism, and making Siwu Decoction whitening cream. Thereby provide research idea and theoretical basis to developing and promoting Siwu Decoction cosmetic effect. The key results are shown as below:1) Using uniform design(based on the table U12*(1210) of uniform design), with Angelica sinensis Radix, Chuanxiong rhizoma, Paeoniae radix Alba and Rehmanniae radix Praeparata of Siwu Decoction as study factors,and whitening effect as an indicator to inspect medicine ratio of Siwu Decoction. The results showed that whitening effect of water extract Siwu Decoction mainly works by Rehmanniae radix Praeparata, the greater the amount of Rehmanniae radix Praeparata and the greater whitening effect; and whitening effect of alcohol extract Siwu Decoction is mainly related to the interaction of Angelica sinensis Radix and Rehmanniae radix Praeparata. The fewer interaction between Angelica sinensis Radix and Rehmanniae radix Praeparata(fewer doses), the greater whitening effect.2) Comparing the whitening effect of Siwu Decoction that extracted by alcohol or water. The results showed that ethanol extracts of Siwu Decoction whitening effect better than water extracts, and the heat reflux extraction by ethanol determined as the best extraction method of whitening Siwu Decoction. Using orthogonal experiment design(based on table L9(34), research on three factors of ethanol concentration, extraction times and extraction time. Results showed that the optimum extraction condition is extracting two times for each 1h by 95% ethanol.3) With whitening effect and dry extract yield as the index,studying the refining condition of Siwu Decoction.The refining Effect is better when the whitening effect better and the dry extract yield fewer. The results showed that the static macroporous resin adsorption method is the better method which significantly reducing the impurities of Siwu Decoction, and retaining a large number of active ingredients. The optimum adsorption ratio of liquid to wet macroporous resin is 4 to 1(m L:g).4) To studied several alcohols cosmetic raw materials’ s solubility of whitening Siwu Decoction extract,and to studied the stability of whitening Siwu Decoction extract which made from these cosmetic raw materials. Results showed that 1,3-butanediol was the best solvent of whitening Siwu Decoction extract,and the whitening Siwu Decoction extract per milliliter contains 250 mg crude drugs.5) Based on an experienced creams formulation, optimizing two systems of emulsifiers and thickeners, and studying to the amount of Siwu Decoction whitening extract. Results showed that the best emulsifier is M202 and AVC complex, the best thickener is EG and AVC complex, and the amount of whitening Siwu Decoction extract is 1-3%(the best was 3%).6) To investigate respectively effect of whitening Siwu Decoction on monophenoloxidase(MPO) and diphenoloxidase(DPO) of tyrosinase. Results shows that whitening Siwu Decoction on MPO without suppression, but on DPO with reversible inhibition. Defining the mechanisms of the Suppression type of whitening Siwu Decoction on tyrosinase by Lineweaver-Burk Plot. Results shows that whitening Siwu Decoction is a type of Non-destructive inhibitor of tyrosinase, competing with substrate(alternative Dopa) which belongs the mixed reversible inhibition. The IC50 of the whitening Siwu Decoction on tyrosinase is 250 mg·m L-1.
