Study On Extraction And Activity Of Peony Seed Shell Oil

Peony seed shell is a by-product produced in the process of preparing peony seed oil.With the extensive application of peony seed oil,a large number of peony seed shells have been generated,and the use of peony seed shell oil can further improve the potential value of oil peony.In this study,peony(Paeonia ostii T.Hong et J.X.Zhang)was used as the raw material.Ethanol was used as the solvent,and the peony seed shell oil was extracted by the water bath reflux method,ultrasonic method,and microwave method.The antioxidant and antibacterial activities of peony seed shell oil were explored.1.Study on extraction technology of peony seed shell oil.Peony seed shell oil was extracted by the water bath reflux method,ultrasonic extraction method,and microwave extraction method.In the optimal process of the water bath reflux method:the solid-liquid ratio is 1:10 g·mL-1,the extraction temperature is 90?,and the extraction time is 4 h.The oil content of the peony seed shell reached 24.68±0.67%,and the yields of the oil and non-oil parts were 5.16%and 19.52%,respectively;The optimum conditions for ultrasonic extraction are ultrasonic power of 480 W,the solid-liquid ratio of 1:8 g·mL-1,the ultrasonic temperature of 60?,and the ultrasonic time of 45 min.The extraction rate reached 23.39 ± 1.17%,the yield of oil and non-oil parts were 4.11%and 19.28%respectively;The optimal conditions for the microwave extraction method are the solid-liquid ratio of 1:10 g·mL-1,the microwave power of 560 W,and microwave time of 15 min.Through verification tests under optimized conditions,the extraction rate reached 26.98± 1.63%,the oil content was 5.37%,and the non-oil content was 21.61%.Comparing the three extraction methods,the peony seed shell oil content obtained by microwave extraction is higher.Therefore,this method is selected for extraction.2.Analysis and research on the composition of peony seed shell oil.It is found through research that peony seed shell oil contains sugar,protein,and other nutritional ingredients;?-linolenic acid,total phenol,total flavonoids,paeoniflorin,resveratrol,and other active ingredients.According to the literature,?-linolenic acid has an effective effect in the treatment of malignant tumors,coronary heart disease,and other diseases;resveratrol,paeoniflorin,total phenols,and total flavones have important biological properties in anti-inflammatory,antioxidant,and anti-tumor Features.This proves that peony seed shell oil has significant economic and medicinal value in practical production.3.Study on antioxidant and antibacterial activities of peony seed shell oil.Antioxidation experiments show that peony seed shell oil has scavenging ability to DPPH,ABTS,hydroxyl radicals,superoxide radicals,and liposome peroxidation,especially the effect of scavenging ABTS free radicals;and peony seed shell oil effectively promote the increase of CAT and SOD,and inhibit the generation of MDA.Bacteriostatic experiments showed that peony seed shell oil had good antibacterial effects on Staphylococcus aureus.Therefore,peony seed shell oil be utilized in the development of antioxidant and antibacterial products.
