Porphyrins and phthalocyanines are closed and continuous 18? electron conjugated systems with high thermal and chemical stability.Their unique optical,electrical,and catalytic properties are often used as photosensitizers for tumor diagnosis and therapeutic studies and enzyme-like catalysts.In this thesis,photothermal conversion particles based on alkylamino substituted phthalocyanines have been constructed and their photothermal properties were investigated.Furthermore,luminol chemiluminescence systems based on the metal-organic framework material ZIF-90 platform were fabricated and have been used for glucose detection and tumor treatment,respectively.I.Human serum albumin?HSA?-assisted and molecular phthalocyanine?Pc?-based NIR theranostic nanoparticles?NPs?were fabricated,exhibiting good biocompatibility and photothermal properties.Under the irradiation of near-infrared 808 nm laser,HAS-Pc NPs showed high photothermal response and photothermal stability for tumor photothermal treatment.II.The Fe Pc-lumino ZIF-90 nanomaterial was prepared for glucose chemiluminescence detection.Host-guest nanomaterial Fe Pc@ZIF-90 was synthesized by in-situ self-assembly;Fe Pc-lumino ZIF-90 nanomaterial Fe Pc@ZIF-90-Lum was prepared by Fe Pc@ZIF-90 a post-synthesis method;as a chemiluminescent sensor,it can be effective utilize the chemiluminescence reaction of hydrogen peroxide and luminol catalyzed by Fe Pc to realize the quantitative analysis of glucose.This work provided the possibility for endogenous hydrogen peroxide to trigger luminescence as a built-in light source for photodynamic therapy.III.A self-illuminating ZIF-90-based composite photositive system was fabricated.The obtained PME@Fe-ZIF-90-Lum consists of Fe-doped ZIF-90 host,protoporphyrin IX dimethyl ester?PME?photosensitizer guest and covalently grafted luminol light emitter.Under the given conditions,the light,which is generated from the Fe-catalysed luminol oxidation by endogenous H2O2,can in situ activate the encapsulated photosensitizer PME to generate fluorescence and 1O2 via fluorescence resonance energy transfer?FRET?process.In the absence of external light source,its antitumor photodynamic therapy was successfully realized and its high antitumor efficacy was well evidenced by in vitro and in vivo experiments.These studies provide the possibility for the application of phthalocyanine and porphyrin functionalized nanomaterials in tumor phototherapy and glucose detection.
