Hericium erinaceus is an important medicinal and edible fungus with rich nutritional value.Polysaccharides and oligosaccharides are two important active ingredients.Traditional alcohol precipitation methods have some disadvantages in the separation of polysaccharides and oligosaccharides,such as low yield,large amount of organic reagents consumed,and low sample activity.In this paper,membrane technology was used to study the separation process of polysaccharides and oligosaccharides from Hericium erinaceus,and the key techniques of multi-stage membrane separation were established.The pollution mechanism of polysaccharides and oligosaccharides in the membrane separation process was studied.The content includes the following three aspects:(1)The effects of membrane separation and alcohol precipitation on the separation and antioxidant activity of crude polysaccharides from Hericium erinaceus were studied.The monkey head is taken from the fruit body of the Hericium erinaceus by the hot water leaching and then obtained by the alcohol precipitation,nanofiltration,nanofiltration + alcohol precipitation,microfiltration + nanofiltration,microfiltration + nanofiltration + alcohol precipitation,etc.The crude polysaccharides of the mushrooms were recorded as A-He P,B-He P,C-He P,D-He P and E-He P,respectively.The separation,physicochemical properties and antioxidant activities of crude polysaccharides from Hericium erinaceus isolated by the five methods were compared.The results showed that the separation of microfiltration(PES 0.1 ?m)+ nanofiltration(PA 300 Da)was better.The yield of crude polysaccharide was 10.08%,the purity was 43.01%,and 80.34% of the polysaccharide in the extract was retained.The crude polysaccharide samples obtained by the five treatments contained a certain amount of protein,and the protein content of B-He P was higher than that of C-He P,indicating that the microfiltration treatment can remove some proteins and increase the purity of the polysaccharide.The results of reducing power,·OH,ABTS· and DPPH· clearance showed that the five crude polysaccharide samples had certain antioxidant capacity,and the crude polysaccharides of Hericium erinaceus obtained by microfiltration + nanofiltration process had the best antioxidant capacity.(2)The multi-stage membrane separation process of Hericium erinaceus oligosaccharides(HOS)was established to study the physicochemical properties of HOS and the proliferation of Lactobacillus.The multi-stage membrane process was prepared by extracting the Hericium erinaceus by PES 0.1 ?m microfiltration membrane(MF),PES 1 000 Da ultrafiltration membrane(UF)treatment and PA 300 Da nanofiltration membrane(NF).The HOS obtained by the separation of the process accounts for 16.46% of the total polysaccharide content in the extract,and the purity is 63.71%.Lactobacillus has the best proliferation effect when HOS accounts for 25% of the carbon source.(3)To study the membrane flux,membrane fouling resistance and mechanism of microfiltration,ultrafiltration and nanofiltration in the separation process of polysaccharides and oligosaccharide membranes of Hericium erinaceus.The microfiltration and ultrafiltration membranes mainly form reversible pollution with gel layer as the main,and the nanofiltration membrane mainly forms irreversible pollution mainly by adsorption in the nanofiltration process.The three membranes were best treated by ultrasonic cleaning.The membrane flux recovery rates were 84.61%,82.32%,and 75.08%,respectively,and the porosity increased by 53.24%,45.45%,and 34.79%,respectively.
