A water soluble crude polysaccharide extracted by alkali has been isolated from the fermentative mycelia of Hericium erinaceus Pers. After purified by the combined deprotionization methods of enzyme and sevage, it (HPP) was homopolysaccharide by tests: Gel filtration on Sepharose CL-6B, Cellulose acetate pellicle electrophoresis and Determination of specific rotation. The identification of purificaton implied that HPP was uniform on both molecular weight and polarity, furthmore its molecular weight was about 65.3 thousand by gel filtration, and it was polyglucose by G.C. analysis.Partial hydrolysis with acid, Periodate oxidation, Smith degradation, IR, and Methylation were used on the structure analysis of HPP. After acetylation, the methylated productions were analyzed by G.C.-M.S.The results revealed that HPP was a kind of highly branched structure. The main chain was composed of (1→6) -linked-Glc, and (1→3)-linked-Glc residues were substituted at O-3. That is to say, in every four unit, there was one (1→6)-linked-Glc, together with one (1→6,3)-linked-Glc, which formed the main chain, and there was one (1→3)-linked-Glc, as well as one deoxidized Glc, which made up the side chain.
