Tea saponin has a wide range of biological activities,including inhibition of alcohol absorption,protectionof the gastrointestinal tract,anti-hypertension,anti-inflammation,anti-tumor,anti-oxidation,anti-cough-dyspnea,hemolysis,ichthyotoxin,antibacteria and other effects.Camellia oleifera is one of the world's four main woody oil plants,and is a unique oil plant in China.At present,the research on camellia oleifera mainly focuses on the comprehensive utilization of camellia oleifera seed oil and camellia oleifera cake,and waste utilization of camellia oleifera is rarely carried out.The tea saponin in the camellia oleifera shell has the characteristics of green,harmless and high safety,and has wide application value.As a cash crop widely cultivated in Hainan,the extraction and purification of camellia oleifera provides new ideas for developing a variety of daily necessities.Purpose:the purified tea saponin is extracted from the camellia oleifera shell to do physical and chemical properties testing and activity testing,with the aim of expanding the application route of tea saponin,and to provide a theoretical basis for the comprehensive utilization of the camellia oleifera shell.Methods:the single factor and Box-Behnken experiments are conducted to optimize the extraction parameters of tea saponin and to determine the optimum extraction conditions.The orthogonal experiments are performed to optimize the optimum process for decolorization of tea saponin extract by hydrogen peroxide.AB-8 macroporous adsorption resin and Sephadex G-15 are selected to separate and purify tea saponin.The purified tea saponin is tested for antibacterial activity.Results:the content of tea saponin in camellia oleifera shell is 7.5%.Through the Box-Behnken optimization experiment,it can be seen that the optimum extraction conditions are as follows:the material-liquid ratio(g/m L)is 1:1 2,the extraction time is 6 h,the extraction temperature is 82 C,and the ethanol concentration is 57%.Through the single factor and the orthogonal experiments,it can be seen that the optimum decolorization conditions are as follows:the decolorization time is 60 min,the amount of hydrogen peroxide is 20%,the pH is 10,and the decolorization temperature is 50?.Through the singal factor,it can be seen that the optimum process conditions for determining AB-8 macroporous adsorption resin are as follows:the loading quantity of sample is 1 BV,the loading solution is 25 mg/mL,the loading solution PH is 5,the loading flow rate is 3 BV/h,the elution volume is 4 BV,the elution flow rate is 3 BV/h,the eluent ethanol concentration is 95%,and the eluent pH is 9.Tea saponin with a purity of 82.5%can be obtained by Sephadex G-15.Tea saponin has a good antibacterial effect on both Escherichia coli and Staphylococcus aureus.Tea saponin acts on the cell membrane of Escherichia coli and Staphylococcus aureus,which increases the permeability of the cell membrane,destroys the membrane structure of the cell,ruptures the cell,dissolves the content,and acts as a bacteriostatic effect.
