In order to improve the quality standard of the Rosa roxburghii leaves,the content of the total polyphenols and total flavonoids in the pear leaves was established,and the test solution was prepared based on the antioxidant capacity of different polar parts.To establish the fingerprints of Rosa roxburghii leaves.So as to provide a scientific basis for the stability and quality control of the medicinal materials.In this study,the extraction process of total polyphenols from Rosa roxburghii fruit was studied.The effects of different drying methods on the total polyphenols of Rosa roxburghii fruit were studied.The extraction conditions of total polyphenols were optimized by response surface analysis method,and the extraction of polyphenols Purification technology for the product development to provide a reference.The main research is as follows:1 Improvement of quality standard of Rosa roxburghii leaves?Identify?Perfected the original standard of thin layer identification,with different quercetin as reference substance,polyamide membrane as the stationary phase,an agent for formic acid,ethyl acetate,toluene(1:2:1),chromogenic agent of 3% AlCl3-ethanol solution,the same as the different quercetin position fluorescence spots.?Examination?With reference to the 2015 version pharmacopoeia method for determining the moisture,total ash,acid insoluble ash content,water extract,60% ethanol extract.Tentative limits for each check for moisture less than 9.0%,more than 7.0% total ash,acid insoluble ash content less than 0.6%,the water extract of not less than 26.6%,60%ethanol extract of not less than 32.8%.Using AFS933 non-dispersive atomic fluorescence spectrometry determination of mercury,arsenic,the ZEEnit 700 p type graphite furnace atomic absorption spectrometry determination of lead and cadmium.Heavy metal content is very low,not included in the standard.?Determination of content?The method for the determination of total polyphenols and total flavonoids in Eriocheir sinensis was established by orthogonal experiment design.The content of total flavonoids and total polyphenols were determined.The total contents of total polyphenols and total flavonoids were determined to be 1.48% and11.62% respectively.The limit of total polyphenol content was not less than 8.7% and the total flavonoid content was not less than 1.1 %.?Fingerprint?The preparation of the test Through the thorn pear leaf of different polar parts antioxidant activity comparison(DPPH free radical clearance rate and total antioxidant power as evaluation index),determining n-butyl alcohol layer extracts the strongest antioxidant capacity,so choose the Rosa roxburghii leaves study on fingerprint butanol layer.The chromatographic conditions Phenomenex Gemini 5u C18(110A250×4.6mm)as the column,0.1% phosphoric acid and acetonitrile as mobile phase,gradient elution,wavelength 270 nm,column temperature 30 ?,flow rate 1 mLˇmin-1.The results were analyzed by cluster analysis.The samples were divided into two types according to2015 and 2016.The samples were distinguished from other samples in spring.Five principal components were obtained by principal component analysis.The results of the above results were verified by cluster analysis of the characteristic peaks at 360 nm.2 Study on extraction technology of total polyphenols(1)The study of drying process With total polyphenols as reference factors,decompression drying,atmospheric oven drying and freeze-drying three treatment on the effect of total polyphenols in the thorn pear fruit,vacuum drying and freeze-drying two ways to stab total polyphenol content of minimal impact.(2)Extraction process optimization The single factor extraction and response surface analysis method were used to optimize the extraction process of total polyphenols of Rosa roxburghii fruit.The optimum extraction conditions were as follows: extraction time 48 min,ethanol percentage 60%,liquid ratio 16: 1,The extraction rate of polyphenols in Rosa roxburghii was improved.(3)Using ethyl acetate extraction Rosa roxburghii fruit extract,stab toRosa roxburghii fruit total polyphenols purification process carries onthe preliminary exploration,will stab Rosa roxburghii fruit total polyphenol purity increased from 22.2 % to 41.7 %.
