The interaction between biomolecules can reflect the active function of biomolecules.We study of this relationship is of great significance for the further understanding of the field of life sciences.The surface plasmon resonance imaging(SPRI)technique can detect the interaction between the biomolecules without marking,real-time and high-throughput.In this paper,the development of surface plasmon resonance(SPR)and the research status of SPRI are reviewed.An experimental study on the digital holographic imaging of onion inner lamellar cells was carried out by using a self-assembled digital holographic microscopic imaging device.The holographic image is obtained,and the intensity and phase images of the sample are reconstructed.A surface plasmon resonance imaging device based on digital holographic is assembled by combining digital holographic with surface plasmon resonance.A series of related experiments are carried out by using this device.The evaporation process of 0.2 ?l deionized water on gold film was studied.A total of 202 holographic images were obtained,and the intensity and phase information of water droplet evaporation were obtained.The experimental results are similar to the theoretical simulation.The feasibility of this method and the stability of the self-assembly device are proved.The biochip was prepared by monolayer technique,and the related experiments of drug research and immune detection were carried out.The interaction process of three different concentrations of samples was detected on the same biochip at the same time.The interaction between different molecular weight peach gum polysaccharides(PGP-1,PGP-2)and galactose lectin-3 was detected.And the interaction of glucagon-like peptide-1 derivative(6KTP)with mouse albumin and monkey albumin was detected.Reaction kinetics parameters were obtained and standard curves were established.In addition,the immune response of acidic fibroblast growth factor(aFGF)antigen and antibody was detected by this apparatus.Through this detectionsystem,we can intuitively see whether there is immune response to aFGF antigen and antibody.The immune response process of the sample was monitored in real time without labeling the sample.The experimental results of this paper show that the experimental device and method are simple and feasible,do not need marking,need less samples and low cost,and have good application value in the detection of high-throughput biomolecule interactions.
