ObjectiveIn this study,bioinformatics analysis of gene chips related to non-small cell lung cancer was carried out,and potential key genes in non-small cell lung cancer were preliminarily explored,and the possible signaling pathways and clinical significance of key genes were discussed in depth.Methods1.Download chip data and platform files numbered GSE19804 from high-throughput gene expression database(GEO).2.Quality control check for the level of homogenization of the original data.3.The original data are pre-processed such as background correction and normalization processing.4.The differential genes were screened by the obtained expression matrix,and thermal maps and volcanic maps were drawn.5.On-line tool DAVID was used for functional annotation and pathway enrichment analysis of differentially expressed genes.6.The protein-protein interaction network analysis was constructed by using STRING database and Cytoscape software,and then the key node genes and hub modules were screened out.7.The differentially expressed genes were verified by Oncomine database,and the survival curves of the key genes were obtained by using the(TCGA)of cancer genome map.Result1.A total of 818 differentially expressed genes were screened out,including 561 up-regulated genes and 257 down-regulated genes.2.In the analysis of cell components,down-regulated DEGs were mainly enriched in extracellular space and extracellular region,and up-regulated DEGs were mainly enriched in the surface of cell membrane and extracellular matrix.In the analysis of molecular function,down-regulated DEGs mainly are concentrated in protein binding,protease catalysis and other functions.Up-regulation DEGs mainly are concentrated in cytokine binding,receptor binding and other functions.In the analysis of biological processes,down-regulated DEGs are mainly enriched in mitotic cell cycle,extracellular matrix decomposition,cell apoptosis,cell proliferation and other processes.Up-regulated DEGs are enriched in leukocyte migration,inflammatory reaction,angiogenesis and other processes.3.In signal pathway analysis,down-regulated DEGs mainly involve in extracellular matrix-receptor interaction,cell cycle,PI3K-Akt signal pathway,p53 signal pathway and other related pathways,and up-regulated DEGs are releated to TNF signaling pathway,Cytokine-cytokine receptor interaction,cellular adhesion molecule(CAMs),cancer transcription disorder,NF-kB signal pathway and other pathways.4.Through the PPI network analysis of differential genes,eleven key node genes and three important hub modules were screened.5.The expression of key differentially expressed genes CAV1 and CDH5 in NSCLC was verified in Oncomine database.The survival analysis based on TCGA database revealed that the overexpression of CAV1 and CDH5 might be a risk factor for the prognosis of NSCLC.ConclusionThis study shows that the overexpression of core genes CAV1 and CDH5 was associated with poor prognosis of non-small cell lung cancer.It provides a theoretical basis for the establishment of related clinical prediction models and provides a new target for targeted therapy of non-small cell lung cancer.
