Objective: To explore the expression of nicotinamide N-methyltransferase(nnmt)in non-small cell lung cancer(NSCLC)and its relationship with clinicopathological factors,and to analyze its related signaling pathways by bioinformatics analysis.Methods: Collected 73 cases of surgically resected NSCLC cancer tissues,paired adjacent tissues and corresponding clinical data from the Thoracic Surgery Department of Guizhou Provincial People's Hospital.Immunohistochemical staining was used to evaluate the expression of NNMT in NSCLC cancer tissues and normal tissues adjacent to the cancer,and to explore the relationship between NNMT expression and the clinical pathological characteristics of patients..In addition,the top10 genes closely related to NNMT were studied using the String database,and the signal pathways therebelted in the preliminary discussion is initially discussed based on Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis.Results: There was no significant difference in NNMT expression in lung cancer tissues of different genders,ages and tumor pathological types,and the difference was not statistically significant.NNMT expression may have a certain correlation with tumor TNM staging.With the increase of staging,the positive expression rate of NNMT gradually increases.The expression rate of NNMT is higher in patients with lymph node metastasis,and the difference is statistically significant(P<0.05);the expression rate of NNMT was higher in patients with distant metastasis,but the difference was not statistically significant due to the small number of cases;NNMT expression may have a certain correlation with tumor size,and the positive expression rate of NNMT is higher in patients with larger tumor size,The difference was statistically significant(P<0.05);In addition,10 groups closely related to NNMT are constructed because of AOX1,BST1,CD38,PNP,NAMPT,NNMT,S100A10,AKAP12,FHL2,EGFR,PTRF,and participate in nicotinate and nicotinamide metabolism,salivary secertion,pancreatic secertion,oxytocin signaling pathway,JAK-STAT signaling pathway and other biological pathways.Conclusion: The expression of NNMT in NSCLC cancer tissues is higher than that in adjacent tissues;NNMT expression has nothing to do with gender,age and tumor pathological type,but is related to TNM staging,tumor size,and distant metastasis;NNMT may be involved in several key cancer-related signaling pathways,affecting the occurrence and development of NSCLC,and is a potential biomarker.
