Lung cancer is currently the malignant tumor with the highest morbidity and mortality in the world,which seriously threatens human health and life safety.Yunnan Xuanwei is one of the regions with the highest incidence of lung cancer in my country,and has unique regional epidemiological characteristics.Due to the high malignancy of Xuanwei lung cancer and the lack of effective molecular standards,most Xuanwei lung cancer patients were already in the advanced stage of cancer when they were first diagnosed and missed the best time for treatment.Therefore,it is extremely urgent to study the pathogenesis of Xuanwei lung cancer.However,at present,research on the molecular mechanism of Xuanwei's lung cancer development is very limited,and many important genes have not been discovered.Therefore,the identification of differentially expressed genes unique to the Xuanwei region is of great significance for the research and development of diagnostic biomarkers and the disclosure of the molecular mechanism of lung cancer development.In this study,high-throughput transcriptome sequencing(RNA-Seq)was used to compare the m RNA of the cancer group and the adjacent group in Xuanwei lung cancer,and the bioinformatics method was used to analyze the m RNA of non-Xuanwei lung cancer(other regions of China).Through integrated analysis of flux transcriptome data,by comparing the differentially expressed m RNAs obtained from Xuanwei lung cancer and non-Xuanwei lung cancer,key genes related to the onset of Xuanwei lung cancer were obtained,and through differential functional clustering,signaling pathways and The interaction network between the corresponding protein members.Through relevant literature search on each differentially expressed gene,we selected 6 candidate genes(APLN,GPX3,DLL4,ITGB8,ABCB1,SULF1)and verified their expression levels in Xuanwei lung cancer samples by q RT-PCR,and the results Both are consistent with m RNA sequencing results.In addition,we use the ROC curve to evaluate the clinical diagnostic value of candidate differentially expressed genes.The results show that,except for ITGB8,APLN,GPX3,DLL4,ABCB1,and SULF1 have important diagnostic value for Xuanwei lung cancer.In conclusion,this study conducted an integrated analysis of high-throughput transcriptome data from patients with lung cancer in Xuanwei and non-Xuanwei areas,and conducted a preliminary screening of key genes of Xuanwei lung cancer,which helped to understand the genetic molecules of Xuanwei lung cancer and their regulation.The mechanism provides a certain basis for the diagnosis and in-depth study of Xuanwei lung cancer.
