Objective:Small cell lung cancer(SCLC)accounts for 10% to 15% of all lung cancer patients.Due to high malignancy and early metastasis of SCLC,the patients often have a poor prognosis.This study aims to screen and explore the mi RNAs and genes involved in the regulation of SCLC metastasis and the progression of SCLC,and to predict the targeted drugs that might render therapeutic potential,so as to enrich the mechanism of metastasis in the disease.Methods:Screening mi RNA,differentially expressed in patients with extensive stage versus limited stage by bioinformatics methods to screen and verify genes that may be involved in tumor metastasis and progression by predicting targeted genes and drugs,functional enrichment analysis,construction of protein interaction network and prognostic analysis.The wax slices of tumor and paracancerous tissues of 3 patients with SCLC were selected from the Department of Pathology of Hunan people's Hospital,and the expression of target gene m RNA in SCLC tissue was determined by quantitative PCR.Results:Five mi RNAs that might be associated with SCLC metastasis,42 key module genes,and 16 core genes that were significantly related to poor prognosis in SCLC patients were screened,among which E2F1 and GRB2 had higher mutation potential in patients with SCLC.The results of Q-PCR showed that the expression of E2F1 m RNA in tumor tissues of SCLC patients was higher than that in paracancerous tissues,while the expression level of m RNA of GRB2 in tumor tissues was lower than that in normal paracancerous tissues.56 targeted drugs that might render a therapeutic potential were also predicted.Conclusions:E2F1 and GRB2 may be closely related to the occurrence and progression of SCLC tumor,and this study may provide theoretical basis and research direction for exploring the mechanism of SCLC metastasis,early identification and prediction of biomarkers.
