In order to explore the effect of environmental pH on the protein content of Suillus luteus,Itraq quantitative labeling technique and tandem mass spectrometry were used to sequence and analyze the proteome of the fungus under different pH conditions.Compared with the mycelium proteome cultured under the pH 3.0,there were 497,735,1183,1581,1883,2578,2588 differential proteins respectively,which were annotated to 90,58,65,67,59,44,36 functions in the GO database respectively,and positioned to 34,22,17,18,17,22,16 metabolic pathways in the KEGG database.The content of succinyl-CoA synthase,succinate dehydrogenase,aconitate hydratase,a-ketoglutarate dehydrogenase complex,isocitratc lyase and aconitase is higher under the condition of pH 6.0.The high content of acetyltransferase,citrate synthase and malate synthase promotes the production of citric acid and malic acid under the condition of pH 8.0.Exocyst protein,small GTPase Cdc42,1,3-?-glucanyltransferase,guanine nucleotide-binding protein and nucleolar GTP-binding protein are related to signal transduction,transporting ions and other functions,the high content of which is an important factor to promote the rapid growth of hyphae under the conditions of pH 4.5 and 6.0.Some relatively exuberant metabolic pathways are also one of the reasons for the large biomass of hyphae,such as pyruvate metabolism,fatty acid metabolism,glutathione metabolism,glycolysis pathway,tyrosine metabolism,glycine,serine and threonine metabolism and so on.
