Mycorrhizal fungi play a crucial role in the restoration and reconstruction of forest vegetation.Inoculation of Ectomycorrhizal fungi(ECMF)has a promoting effect on host plants.Using mycorrhizal fungi and high-efficiency bacteria to replace chemical fertilizer the strong seedlings were cultured.The experiments was conducted on the Pinus tabulaeformis and the P.sylvestris var mongolica inoculated with Suillus luteus.Soil samples were collected.Using Illumina High-throughput Sequencing,BIOLOG techniques,soil microbial diversity and microbial activities were analyzed in order to track the time-lapse after 5 years inoculated with S.luteus.The characteristics of changes in soil microbial communities and diversity were revealed.The results were as follows:1)The water content,organic carbon content,available phosphorus content,and available nitrogen content of the soils of two pines inoculated with S.luteus were higher than the control and the pH value showed a state of decreasing.2)The two pines inoculated with S.luteus significantly increased the use of carbon sources,the richness index and evenness index of rhizospherical soil microbes.However,the dominance index was decreased.3)At the genus level,all treatments soil included RB41,Pseudarthrobacter,Sphingomonas,Bacteroides,Solirobrobacter,H16,Variovorax etc.After the inoculation of S.luteus,the community structure of the P.tabulaeformis and the P.sylvestris var.mongolica were effect and the UPGMA clustering tree demonstrated the same community structure of the two pines inoculated with S.luteus.4)At the genus level,all treatments soil included Geopora,Suillus,Rhizopogon,Inocybe etc.After inoculation with S.luteus,the relative abundance of Suillus and Ilyonectria increased significantly.Moreover,after S.luteus inoculation,the Shannon indexes and Simpson diversity indexes of the microorganisms of the two pines were significantly increased.5)At the genus level,the detected dominant genus included Geopora,Suillus,Rhizopogon,Inocybe etc.After S.luteus inoculation,the relative abundance of Suillus significantly increased,especially in the ECM fungi of the P.tabulaeformis.After S.luteus inoculation,the microbial diversities increased significantly.6)The pH,soil water content(SWC)and available nitrogen(AN)have a great influence on the community composition of soil bacteria,fungi,and ECMF,especially pH values play a key role in the distribution of fungi and ECMF in two pines inoculated with S.luteus.In summary,the inoculation of S.luteus changed the community structure and diversities of soil microorganisms in the two pines.Increased utilization of carbon sources and microbial metabolic activities by soil microorganisms.After five years of inoculation,S.luteus is still dominant.This supports the research and development of mycorrhizal afforestation.To lay the foundation for the further development of microbial agents that utilize mycorrhizal fungi.It has important theoretical value and practical significance.
