Suillus luteus is a more extensive distribution of ectomycorrhizal fungi, with the Pinaceae plant symbiosis, improve soil environment, increase the uptake of water and nutrients. The optimum pH value of its growth was 5.5, when the optimum pH value higher, Suillus luteus reduce the environmental pH value through the production of oxalic acid and other organic acids. And the higher the environmental pH value, the more the amount of oxalic acid. In order to clear Suillus luteus how to sense the the pH information, secrete oxalic acid and adjust the optimum pH. We study the effects of Na3VO4 and CuSO4 on plasma membrane H+-ATPase activity and the influence of mycelial biomass, pH value variation and oxalic acid production. The results showed that the activity of H+-ATPase was higher in the range of pH6.0~7.0, and the optimum pH value was 6.5. H+-ATPase activity in low concentration of Na3VO4 and CuSO4 have an increasing trend, and then with the increase of the concentration, the enzyme activity gradually decreased. Na3VO4 inhibitit the plasma membrane H+-ATPase activity, at the concentration of 200μmol/L, activity is inhibited about 67.3%. CuSO4 inhibitory effect on the plasma membrane H+-ATPase activity is weak, at the concentration of 200μmol/L, only inhibited about 9.5%. Suillus luteus plasma membrane H+-ATPase involved in environmental pH adjustment and oxalic acid synthesis, to adjust and maintain the growth of the hyphae by the mutual coordination of the enzyme activity, pH value and oxalic acid.
