Rumex japonicus Houtt.is a perennial herb belonging to the Polygonaceae, with the character of efficient fertility as well as extreme low living request. In this study we conduct “traditional Chinese medicine fingerprint” to identity the accumulating regulation of three main secondary metabolites including e modin, chrysophanol and rheochrysidin of R.japonicas exposed to natural env ironment(Three sampling points: YN-the small yard in Life Science Buildin g of Jilin University,NW-the bush near No.5 Nanyuan Dormitory of Jilin U niversity,XH-the grassland lying by the west gate of Jilin University)and i ndoor salt-alkali environment(A1-A5: saline environment,B1-B5: alkaline env ironment,CK: control group)in order to work out the environmental factors of accumulation. Our results are shown as following:1.The accumulating regulations of R.japonicus in different living environ ment.(1)Root: The three main secondary metabolites in roots perform a same tr end where higher accumulation comes to YN and NW sampling points compa ring with XH.(2)Shoot: The regulation of three contents defers from each other. The e modin in YN and NW sampling points shows numerous accumulation of seco ndary metabolites, following with a low build-up tend in XH.As for the chr ysophanol,it manifests a peak in NW sampling point during the vegetative p eriod(seeding period to bolting period)and then turns to another high point in XH sampling point during the productive period(flowering period to fruiti ng period). When it comes to the rheochrysidin, high level of accumulation o ccurs in seeding period and flowering period, the other three period encounter lower standards.2.The accumulating regulations of R.japonicus in every living environmen t under different living phases.(1)Root: the three components get into the same trend where it rises up i n seeding and growing periods, lows down in bolting period and then underg oes towards a peak in flowering, finally, diminishes to a low ebb.(2)Shoot: The regulations carry out different strategies, the content of the emodin and rheochrysidin are totally opposite: when the one rises up,the ot her goes down.In addition, the content of chrysopanol, the tendencies in thre e sampling points get the same of raising up at first and follow with a fall. However, the peak of chrysopanol is not always the same point,it distinct as the environment alerts: YN-bolting period,NW-breeding period,XH-flowerin g period.3.The accumulating regulations of R.japonicus under saline and alkaline c onditions.(1)Neutral-salt(saline)environment: In roots the content of secondary met abolites keeps increasing in saline environment, since two concentration group(A4 and A5)where the accumulations get the top are pointed out;while in shoots, with the treatment of salt the content of three main secondary metabo lites undergoes a low tend at first, then an up-going tendency occurs.Overall, the peaks comes to A1 as well as A5.(2)Basic-salt(alkaline)environment: The content of three main secondary metabolites in roots accumulates in B3 or B4 but decreases in a higher conce ntration group(B5),while in shoots the content suffers from a lift in B2 or B3 following with a decline in higher concentration groups.4.Factors that affecting the accumulation of three main secondary metabolites in R.japonicus.(1)Outdoor natural environment: The accumulation of major secondary me tabolites in is relevant to its biomass while in shoots the factors which contri bute to build up the secondary metabolites lie in not only biomass of shoots, but also the stem height, water content, photosynthesis and chlorophyll.Addi tionally, only the former two indexes make big contributions.(2)Indoor salt-alkali environment: In roots, the morphology(root length and root thick),organic acid(Proline, soluble sugar and MDA)correlate with themajor secondary metabolites, meanwhile, both of the two indexes show great influence in component accumulation. As for the shoots, photosynthesis(photo synthetic rate, transpiration rate and stomatal conductance)makes sense.
