The study of intestinal microbiota is one of the most popular research contents in recent years.Intestinal microbes are the most abundant microorganisms in the human body,and the genomes they contain can be called"the second human genome".With known physiological functions such as nutrient absorption,metabolic regulation and immune regulation,it also plays a vital part in life activities.Since the US National Institutes of Health launched the "Human Microbiome Group Project",and the European Union launched the seventh project of the gut genome project,many discoveries have been obtained.A large number of studies have indicated that many diseases such as intestinal diseases,type 2 diabetes,obesity,depression,multiple types of cancer,and excessive immune system activity are associated with intestinal microbes.Hence,this line of research may provide a new avenue for treating these diseases.Traditional Chinese medicine is a gem that has been circulated for thousands of years by the Chinese nation.Its various active ingredients can cure a number of diseases or regulate body functions.For intestinal microbes,polysaccharide components play an important role in maintaining the balance of intestinal microflora.Traditional Chinese medicine can regulate intestinal microbes by adding probiotics and reducing harmful bacteria.However,the specific process remains to be further explored.At present stage,due to the few species of microorganisms that can be cultured in vitro,research on intestinal microbes mainly relies on the extraction of metagenomic DNA and high-throughput sequencing technologies.This study compared three different fecal bacterial metagenomic DNA extraction methods,comprehensively taking various factors into consideration,such as experimental requirements,DNA extraction effects,simplification of operation,and eventually chooses a suitable method for the metagenomic DNA extraction of subsequent experimental sample.The potential effects that two traditional Chinese medicine extracts Galla chinensis,Terminalia chebula,and a pure compound gallic acid have on the population of intestinal microbes are studied by continuously intragastric administration to the mice.Bst DNA polymerase large fragment is used to the isothermal amplification and fluorescent labeling of the 16S rDNA fragments of metagenomic DNA from fecal samples.We attempt to use gene chip to detect and analyze the intestinal flora,break the reliance on the next-generation sequencing,and verify the feasibility of gene chip to detect intestinal flora.The research content of this dissertation can provide reference for the selection of metagenomic DNA extraction methods.It can demonstrate the feasibility,simplicity and timeliness of gene chip detection and analysis of intestinal flora,thus opening up a new direction for research approaches to intestinal microbes.According to the research,Galla chinensis,Terminalia chebula,and a pure compound gallic acid have a certain regulation effect on intestinal microbes.In the future,with application of intestinal flora gene chip to clinical detection and scientific research,compared with the next-generation sequencing,not only the cost will be greatly reduced,but also the efficiency will be significantly improved.
