The F-box protein is part of the E3 ubiquitin ligase SCF complex.Abscisic acid(ABA)is an important plant hormone.Abscisic acid plays a role in seed dormancy,seed germination,root development,abiotic stress response and induction of stomatal closure.Light is one of the most important environmental signals of plants.In order to adapt to environmental changes,plants have developed a variety of photoreceptor systems,including phytochromes,cryptochromes,and phototropes.In order to deepen the understanding of the F-box gene and to search for Arabidopsis F-box genes which regulated by abscisic acid or light,the following analysis and experimental validation were performed in this study:1.Using bioinformatics methods to analysis the GEO gene microarrays,we screened 38 ABA-regulated Arabidopsis F-box genes,14 blue light-regulated Arabidopsis F-box genes,and 19 red light-regulated F-box genes.In these genes,At1g15670,At2g18280 and At2g24540 genes were found in both the blue light-regulated and ABA-regulated Arabidopsis F-box genes.At1g15670,At1g61340,At2g36090 and At5g02990 genes were found in both the red light-regulated and ABA-regulated Arabidopsis F-box genes.2.Through promoter analysis,it was found that the elements of the 38 candidate genes regulated by ABA were mainly related to ABA,drought and stress.Both ABA-regulated and light-regulated candidate genes have light-related elements.3.We screened for genes with the largest change in expression and performed quantitative PCR experiments on these genes.Six Arabidopsis F-box genes regulated by ABA,four genes regulated by blue light,and two genes regulated by red light were screened out.The real-time quantitative PCR analysis of these genes was generally consistent with the results of the gene chips.4.This study analyzed the expression profile of candidate genes.The results showed that some of the candidate genes regulated by ABA exhibited high expression in guard cells,seed coats,pollen and senescent leaves.Most of the genes changed significantly in ABA treatment,stress and water absorption.In addition,the expression profiles of 14 candidate F-box genes regulated by blue light and 19 candidate F-box genes regulated by red light show that these genes are expressed to varying degrees in 10 stages of plant development.The above studies have deepened the understanding of the F-box gene.This helps to explore the role of the F-box gene in the plant's process of abscisic acid or light regulation.This provides new research ideas.
