Protein families that contain domain of unknown function(DUF)are widely existed, they usually comprise large number of members, with uncharacterized function in living organisms. The DUF protein family often contains one or more DUF domains.According to the type of DUF domain, they were divided into different families. The DUF220 protein family is predominantly present in the cruciferous family and may play an important role in plant growth, development and response to stress. So far, there have been few studies of the DUF220 family, and in,Arabidopsis thaliana, only the At1g70480 gene in this family has been reported,to be related to jasmonic acid synthesis. In this study, the transcriptional level of the DUF220 gene family was analyzed using A.thaliana as the material, and the function of At1g70480 in this family was explored.Transcriptional level analysis under stress and the effects of different hormone treatments were carried out separately, and the functions of this gene was preliminarily analyzed, and the results were as follows:1.DUF220 gene family members of A.thaliana are specifically expressed in flowers and pods, which may play an important role in flower and pod development.2.Overexpression of At1g70480 gene in A.thaliana did not affect its response to drought stress and methyl jasmonate treatment.3.GUS histochemical staining of the PAt1g70480:GUS transgenic A.thaliana showed that GUS activity mainly presented in leaves, flowers, both ends of the silique and epidermal hair, and the application of exogenous SA inhibits GUS activity. The above results show that the A.thaliana DUF220 gene family plays an important role in flower and pod development, and the expression of the At1g70480 gene, one of the members of the family,is inhibited by exogenous SA.It provides research materials and preliminary theoretical basis for further analyzing the functions of members of the DUF220 gene family.
