Arabidopsis thaliana is one of the most important model plant. To develop an efficient in vitro culture method is an important task in plant biological studies. We established 6 Arabidopsis thaliana suspension cell lines and 1 callus line of different genotypes, which gave a useful material for the following experiments.Using Arabidopsis thaliana suspension cells, two cryopreservation techniques without programmable freezers were carried out. In the vitrification, the cryoperserved cells achieved a 86.21% survival rate (TTC method). While in the protocol desiccation, this is 3.25 (dry weigh of cells revived 20 days/dry weigh of samples). Furthermore, fertile the plants were regenerated form the revived cells. Cryopreservation, as an appropriate method for long-tune preserving plant source, has developed rapidly over the past decade. In fact, this technique has already become an important field of plant biotechnology studies.Cells of different ABA synthesis or response mutants of Arabidopsis thaliana had different viability after cryopreservation, which revealed ABA has a closed relationship with cold- and dehydration-stress signaling transduction. hi our experiments, mutant abil and abi4 have highest survivals after cryopreserved, which also accorded with their status in stress transduction pathways.Quercetin, genistein and aminophyllin, as the inhitory or stimulatory regulator of ion channel, were applied in pre-culture steps in desiccation. They can regulated the activation of ion channel, then acted in the cell stress signaling, changed the tolerance of cells. Single application of quercetin, genistein or aminophylline are negative factor to ABA signaling, but combinative application of quercetin and aminophylline, appeared enhance the efficiency ofABA signaling transduction.
