A variety of pathogenic factors and inflammatory cells in the lungs caused by a large number of accumulation and infiltration,excessive expression and release of inflammatory medium,etc.,make the pulmonary capillary permeability increase in Acute Lung Injury(Acute Lung Injury,ALI).In particular,during the outbreak of new avian influenza epidemics such as SARS and H7N9,severe stress lung injury is the cause of death of most infected people.The dialectic of Chinese medicine,SARS,H7N9 and other infectious diseases is the "epidemic" syndrome.According to the different stages of "wei,qi,ying,xie",the early application of the prescription of qing fever detoxification formula is very urgent.ALI takes place in the critical stage,belongs to the blood of the disease camp,the hot poison goes deep into the blood,the treatment should be based on clear heat detoxification,diarrhea and cold blood.Qingwen Baidu Decoction has the effect of clearing "qi,xie" heat,and it is full of heat and blood.Regardless of ancient and modern it's as a conmon clinical treatment of infectious diseases("epidemic "syndrome)prescriptions.ALI has a rapid onset,difficult to control the course of disease,and the clinical lack of rapid and effective prevention and treatment of drugs.Qingwen Baidu Decoction as a new approach is provided for the discovery of drugs for clinical efficacy.Therefore,this topic is intended to seek effective treatment drugs from the Qingwen Baidu Decoction.According to the characteristics of the medicinal effect,the combination of the disassembled side and the best removal method are selected.Partial least-squares regression method was used to analyze the correlation of"spectrum" and "effect" of the best disassembly,to obtain the pharmacodynamic components,in order to find the effective treatment drugs of ALI,and to guide the clinical rational drug use.The research contents of this thesis are as follows:The first part is the study on the compatibility of the disinfectants and the selection of the best group.In group preliminary optimization of heat-antipyretic disease and disassemble the compatibility study of parallel compare different demolition party group intervention pharmacological indexes of acute lung injury in rats,in order to select the best split party group efficacy material base to study further.The results of the study showed that the number of PMNs and WBC in BALF in the group C1,C2 and C3 were significantly reduced(P<0.05),and the results were the best in C3 group(P<0.01).The protein concentration of C1,C2 and C3 was significantly decreased(P<0.05),with the most significant(P<0.01)in C2 group.TNF-alpha in C1,C2 and C4 groups was significantly reduced(P<0.05),with the best of C1 group(P<0.01).The W/D values(P<0.01)were significantly reduced in each group,with the most significant(P<0.01)in the C3 group.It can be seen that the "clear and hot diarrhea"--C1 group,"heat and cold blbod"--C2 group and "heat and dry dampness"--C3 group can significantly improve the pathological state of ALI in rats induced by LPS.Most scholars believe that excessive,out of control of inflammation is the primary cause of acute lung injury caused by various causes,more performance for neutrophils(PMNs)dependence,regulation of pro-inflammatory and anti-inflammatory factors tend to be dynamic balance is fundamental for the treatment of ALI.Therefore,this topic chooses to study the pharmacodynamic material basis of"qingrexiehuo" group first.In the second part,the partial least squares regression method is used to analyze the functional components.In this paper,the pharmacodynamics experiments of different extracts from the"qingxiuhuo" group were carried out in rats with acute lung injury,and the HPLC characteristic spectrum was established simultaneously.By applying the method of partial least-squares regression analysis dismantle square group parallel pharmacological indexes of different extract with the corresponding HPLC multivariate correlation information,find the open party group intervention the effect of acute lung injury in rats into groups.The results showed that the cumulative effect of X4,X6,X9,X14,X16 and X17 on the decrease of leucocyte and neutrophil in BALF of ALI rats was 88.76%,which was assumed to be "functional groiping".The cumulative effect of the decrease of protein concentration in BALF in ALI rats was 84%,which was estimated to be"functional grouping".Peak X3,X7,by 8 X10,X11,X18,X19,X21 influence ALI rats lung wet dry ratio to reduce the cumulative effect of the contribution rate of 85%,speculation as "efficacy into group",further efficacy test and verify.Part ? LC-MS/MS analysis,separation and structure identification.To find out the HPLC map information corresponding to each component in the functional group of rats with acute lung injury in rats,and to conduct le-ms/MS analysis to preliminarily determine the chemical structure of each functional component.In order to further confirm the chemical structure of various functional components,the structure was confirmed by the separation and preparation of various functional components.According to the LC-MS total ion flow chart and the secondary ion fragment map,18 compounds were analyzed in combination with standard comparison and related literature review.White blood cells and intervention of ALI rats neutropenia"active components,peak X4,X6,X9,X14,X16,X17,in order to Deacetylasperulosidic acid,Scandoside methyl ester,Jasminoside B,Jasminoside E,iangiferin and rutin.Separation by HPLC and TLC legal efficacy composition,the rutin,Genpin-1-?-gentiobiside,geniposide,quercet:n,?-sitosterol,Scandoside methyl ester and Crocin-I,a total of seven monomer compounds.HPLC showed that the peak X6 for Scandos:ide methyl este,peak X10 to Genipin-1-?-gentiobiside,peak X13 for geniposide,peak X17 to rutin,peak X19 as Crocin-I,consistent with LC-MS/MS analysis that the results;In addition,the efficacy of the intervention in the reduction of leucocyte and neutrophilic granulocytes in ALI rats was firther confirmed.
