Traditional Chinese medicine compound has the characteristics of comp lex components, multi way, multi targets and so on, these features make it m ore challenging to study the pharmacodynamics and pharmacokinetics compar ed with chemical medicine. Therefore, it is the key way to establish suitab le pharmacokinetic and pharmacodynamic methods of traditional Chinese med icine in this field. And the concepts of spectrum dynamics and spectrum ef ficiency are put forward, which provide a new research idea for researcher s.Purpose:Taking Sini decoction as model drug, and using the Total Quantum Stati stical Moment (TQSM)to study the spectrum dynamics for pharmacokinetic pa rameters for multi components of Sini decoction and then to study pharmac odynamic of myocardial ischemia model with the grey relational analysis.0 n the basis of fingerprint of traditional Chinese medicine, we explore the spectrum efficiency relationship for multiple components of Sini decocti on on the anti myocardial ischemia effect, and it provides a new method fo r the study of metabolism mechanism and the basis of effective substance of traditional Chinese medicine compound.Methods:1 Establishment of fingerprint for Sini decoctionTo investigate the chromatographic conditions for Sini decoction by t he optimization of flow phase, column, elution gradient, wave length, velocit y and so on;To establish fingerprint of 10 batches Sini decoction and mea nwhile do the similarity researches by " Similarity evaluation system of c hromatographic fingerprint for TCM 2004 revision A";The contents of hypa conitine, mesaconitine, benzoylmesaconine, glycyrrhizic acid, glycyrrhetinic acid and 6-gingerol were determined under the condition of fingerprint ch romatogram of Sini decoction.2 The study on the spectrum dynamics of Sini decoctionRats plasma fingerprints of different time points in the blank and af ter administration were established, with the analysis by the Total Quantu m Statistical Moment, in order to calculate the total statistical paramete rs of Sini decoction and study the spectrum metabolism characteristics of Sini decoction in vivo in SD rats.3 Pharmacodynamic study on anti myocardial ischemia of Sini decoction104 SD rats were randomly divided into 4 groups,8 rats each group, inc luding control group, model group, Sini decoction groups of 10 batches, posi tive control group, with gavage administration for 7 days, from the fifth d ay, except blank control group, rats of all other groups were injected ip i soproterenol (ISO) preparation to make myocardial ischemia model, the cont ents of creatine kinase (CK) and lactate dehydrogenase (LDH) were used as the detection indexes for investigation of anti myocardial ischemia effe ct of 10 batches of Sini decoction samples.4 The study on the spectrum efficiency of Sini decoctionCombined with fingerprint data, the contribution of Sini Decoction on the effect of myocardial ischemia was calculated by the method of grey co rrelation analysis.Results:1. Determination condition for the fingerprint of Sini Decoction was t hat chromatographic column:Agilent C18 (2) (250×4.6mm,5μm);mobile phase: acetonitrile-0.1mol/Lphosphoric acid;elution gradient:(0~18min,5%~19% A; 18~42min,19%~22%A; 42~52min,22%~ 35%A; 52~80min,35%~65%A;80~ 85min,65%~65%A);flow rate:0.8mL/min;detected wavelength:230nm;column temperature:30℃. Fingerprints of 10 batches of Sini Decoction were establ ished and 22 commom peaks were found in Sini Decoction fingerprints. The s imilarity of 10 batches was all over 0.9. This method has good stability, r epeatability and specificity. The contents of hypaconitine, mesaconitine, be nzoylmesaconine, glycyrrhizic acid, glycyrrhetinic acid and 6-gingerol in S ini Decoction were 13.01 ~ 13.03,20.2~20.4,23.1~23.2,364.7~365.8,680. 7~681.3,61.2~62.2μg· mL-1 in 3 batches.2. The metabolism apparent half-life of Sini Decoction was 9.383h in r at in vivo, and it metabolized 95% though 9.383+1.96×15.04h after admini stration.3. Pharmacodynamic experiments showed that compared with the control g roup, CK and LDH in serum of rats in model group were significantly increa sed. Compared with the model group, Sini decoction treatment groups can dec rease CK, LDH contents in serum of myocardial ischemia rats significantly. 10 batches of Sini decoction have good anti myocardial ischemia effect.4. Related parameters of spectrum efficiency were obstained by gray co rrelation analysis, preliminary determination of size order of each common peak against myocardial ischemia was P12>P17>P11>P18>P4>P10>P16>P20>P2>P 15>P14, and the components of number1,3,4 respectively were benzoylmesacon ine, glycyrrhizic acid and 6-gingerol.Conclusion:The spectrum dynamics and spectrum efficiency methods used in this st udy can be applied to study the pharmacokinetics and pharmacodynamics of the traditional Chinese medicine compound, it can reflect the integrity of .the Chinese traditional medicine and provide new ideas and experimental bases for the modernization of traditional Chinese medicine.
