| Objective;The purpose of this study is to evaluate the efficacy of chemical thrombocytosis and its main ingredient,zedoary oil,in the treatment of cervical HPV infections in vitro cytology,molecular biology,and clinical efficacy observations,and to explore its intrinsic mechanism of action for the prevention and treatment of cervical cancer by Chinese medicine to provides research basis.Method:1.Clinical research:42 cases of cervical HPV infection were randomly divided into treatment group and control group of 21 cases.The treatment group was treated with phytotoxicity suppository.One capsule per night was placed in the vagina.After 3 days of menstruation,the patient was continuously treated for 2 weeks.2 weeks was a course of treatment.After the interval of 2 weeks,the second course of treatment was performed.The total treatment was 3 a course of treatment The control group was treated with Baofukang Suppository,2 capsules per night,and the drug was placed in the vagina.After 3 days of menstruation,it was continuously treated for 2 weeks,2 weeks was 1 course of treatment,and 2 weeks later,the second course of treatment was performed.The patient needed a total of 3 courses of treatment.The patient stopped taking the drug during menstruation.Patients were reexamined after three days after the end of the fourth month.Through the comparison between the two groups,and Comparing before and after treatment in a group,we will analyze the change of clinical symptom scores before and after treatment and the condition of negative HPV,so as to evaluate the clinical efficacy of Huadu Suppository in the treatment of cervical HPV infection.2.Experimental study:Measurement of Cell Culture and Cell Proliferation Curvature:HeLa cells were cultured in vitro and each was added with zedoary turmeric oil and the same volume of blank solvent(DMSO).The scorpion cells set up 3 parallel controls.We will detect the effect of Zedoary oil on Hela cell colonization by MTT assay.The expression level of Survivin mRNA in cells was detected:Quantitative analysis of Survivin gene mRNA in Hela cells of blank control group,interferon group and zedoary oil group was performed using quantitative fluorescence assay with GAPDH as an internal reference.Results:1.Clinical study:1.1 Forty-two patients with cervical HPV infection were randomly divided into treatment group and control group with 21 cases each.The treatment group used Huoxue detoxification and dampness as the method.In the treatment group,Huoxue detoxification and dampness were used as the method.After treatment with Huadu thrombolytic therapy,HPV was negative in 12 patients.The negative rate of HPV was 57.14%.Among the results of clinical symptom scores,21 were included in the treatment group,1 was cured,11 were markedly effective,6 were effective,and 3 were ineffective.The total effective rate was 85.71%;According to the results of TCM syndrome scores,the treatment group included 21 patients,5 recovered,6 markedly effective,9 effective,and 1 ineffective.The total effective rate was 95.24%.The clinical symptom scores and TCM syndrome scores before and after treatment in the treatment group were statistically analyzed and the P values were all less than 0.05.The difference was statistically significant.After comparing the changes of the main symptoms before and after treatment,the amount of leucorrhea,color,taste,quality,and lumbar and abdominal pain,abnormal urine,tongue and veins were all improved to varying degrees,of which the secretion was most improved.1.2 The control group was treated with Baofukang suppository.After treatment,8 cases of HPV were negative,and the conversion rate of HPV was 38.10%.Among the results of clinical symptom scores,21 were included in the control group,1 was cured,6 were markedly effective,8 were effective,6 were ineffective,and the total effective rate was 71.43%;According to the results of TCM syndrome scores,21 were included in the control group,1 was cured,5 were markedly effective,10 were effective,5 were ineffective,and the total effective rate was 76.19%.The P value of the clinical symptom scores and TCM syndrome scores before and after treatment in the control group were all less than 0.05 after statistical analysis,and the difference was statistically significant.After comparing the changes of the main symptoms before and after treatment,the amount of leucorrhea,color,taste,quality,lumbar and abdominal pain,and abnormalities of urine and feces were all improved to varying degrees,of which the secretion was most improved.1.3 Comparison of clinical symptoms before and after treatment between the treatment group and the control group.Statistical analysis showed that P was greater than 0.05,indicating that there was no statistical difference in the improvement of clinical symptoms between the two groups before and after treatment.The comparison between the treatment group and the control group before and after the treatment of the improvement of the TCM syndrome statistical analysis obtained P less than 0.05,and the difference between the treatment group was Zedoary oil can inhibit the proliferation of cervical cancer Hela cells,and its inhibitory effect gradually increases with the concentration within a certain concentration.2.2 At 12 hours of culture,the use of interferon in the control group significantly inhibited the expression of Survivin mRNA.When cultured for 12-48h,interferon can inhibit the mRNA expression of the gene.Within 24 hours of culture,the inhibition of expression increased gradually with time.At 24 hours,interferon inhibited the expression of Survivin mRNA in Hela cells of cervical cancer.2.3 Interfering with HeLa cells with low dose of Curcuma oil,within 24h to 48h of culture,the low-dose zedoary oil significantly inhibited the expression of Survivin mRNA,and its inhibitory effect gradually increased with time.At 48 hours of culture,low dose zedoary oil had the best effect on the inhibition of Survivin gene,and its inhibitory effect was superior to that of interferon at this time point.2.4 The curative effect of zedoary oil on the expression of survivin mRNA in Hela cells was significantly inhibited when the medium-dose zedoary turmeric oil was cultured for 5 and 24 hours.Conclusions:1.According to the analysis of clinical efficacy,it can be concluded that the use of Huoxue Detoxification and dampness as the basic treatment method can effectively block the persistent infection of cervical HPV in patients with wet poisonous accumulation syndrome,and has a significant effect on improving the local response of the cervix,and it can also help Regulates body condition.2.Experimental Study:Zedoary oil can effectively inhibit the growth of cervical cancer Hela cells,and zedoary oil inhibited the expression of Survivin gene in Hela cells.However,the optimal concentration and time for inhibiting cell proliferation did not coincide with the optimal concentration and time for inhibiting gene expression.It was speculated that curcumin oil may inhibit the growth and proliferation of cervical cancer Hela cells by inhibiting the expression of Survivin gene.However,the main mechanism of action of zedoary oil inhibiting Hela cell proliferation is not limited to this target.We need to conduct more in-depth and detailed explorations to discover the mechanism of action of Zedoary Oil. |
