| Climacteric syndrome(CS)happens at 45 to 55 years old,by the growth period of transition to the old age stage,this stage ovarian function decline,the estrogen level has decreased with autonomic nervous function disorder appear menstrual disorders even amenorrhoea,hot flashes,sweating,sleep disorder,paresthesia,mood swings and other comprehensive symptoms.Zi Yin An Shen prescription(ZYASP)is a clinical experience prescription from Qin Yue-hao archiater who is a famous old doctor of traditional Chinese medicine,it is mainly composed of Shu Di,Shan Zhu-yu,Shan Yao,Gou Qi-zi,Tu Shi-ziand so on,with the effect of nourishing kidney Yin,raising five organs and regulating Yin and Yang,used in the treatment of climacteric syndrome,symptoms with menstrual disorders,hot flashes,sweating,irritability,insomnia,heart palpitations and other symptoms,has the good clinical curative effect.For further system evaluation of ZYASP pharmacological action,from the level of the whole animal,this study observed the intervention effect of ZYASP on pathology of climacteric syndrome in animal models and the clinical symptoms of female climacteric syndrome,in order to provide experimental basis for clinical application of ZYASP and research and development of new drug of traditional Chinese medicine.1 The intervention of ZYASP on all indexes of experimental menopause model rats.Objective To observe the effect of ZYASP on experimental menopause model rats and to analyze its mechanism.Methods To establish the menopause rats model by ovary removal method,menopausal rats were randomly divided into the menopause model group,the ZYASP high,middle,low-dose groups,the estradiol valerate 0.00009g/kg group,and the Geng Nian An tablet(GNAT)0.48g/kg group,there was also a sham operation group,each group of 12.For 30 days gavage,to test rats autonomous activities and elevated cross maze,and plasma Follicle stimulation hormone(FSH),luteotropic hormone(LH),Estradiol(E2)and Progesterone(P)in rats were measured by radioimmunoassay,and the thickness of endometrium was measured by light microscope;at the same time,high performance liquid phase electrochemical detection was used to determine the content of dopamine(DA),5-hydroxyindole acetic acid(5-HIAA),5-hydroxytryptamine(5-HT)in the prefrontal cortex and hippocampus of rats.Results Compared with sham operation group,model group rats plasma FSH significantly higher(P<0.01),E2,P decreased significantly(P<0.05),viscera coefficient of the uterus,endometrial thickness decreased significantly(P<0.01);the number of autonomous activities decreased significantly(P<0.05),and the proportion of the number of visits to open arms and the proportion of residence time in the open arms area decreased significantly(P<0.05).The DA,5-HIAA and 5-HT of the prefrontal cortex and the hippocampus were significantly reduced(P<0.05,P<0.01).Compared with model group,the ZYASP high,medium and low-dose group of E2 level in rat plasma,viscera coefficient of the uterine and endometrium thickness has a certain degrree,and the ZYASP high,middle-dose can lower plasma FSH level;in behavioral science,ZYASP high,middle-dose can improve the autonomous activity of rats to some extent,and ZYASP high,medium and low-dose ZYASP can significantly increase the proportion of the number of visits to open arms and the proportion of residence time in the open arms(P<0.01,P<0.05);from the level of neurotransmitter,the high,medium and low-dose of the ZYASP significantly increased the levels of 5-HIAA and 5-HT in the prefrontal cortex(P<0.01,P<0.05);and the high and medium-dose of the ZYASP significantly increased the level of 5-HIAA and 5-HT in the hippocampus(P<0.01).Conclusion The ZYASP can reduce menopausal rat plasma FSH level,improve the level of E2,increase viscera coefficient of the uterine and endometrial thickness,and improve the symptoms of depression and anxiety by raising the brain 5-HIAA and 5-HT levels.2 The sedative hypnosis,anti-depression and antiperspirant effect of ZYASP on normal mice.Objective To observe the effect of mother su granule on normal mouse behavior and normal rat sweat model.Methods The female ICR mice were randomly divided into the normal control group,the ZYASP high,middle,low-dose groups and diazepam 0.001mg/kg group,each group of 12.For 7 days gavage,Intraperitoneal injection of napental(0.03g/kg)was performed,and the number of mice and the latency time of sleep were recorded.The female ICR mice were randomly divided into the normal control group,the ZYASP high,middle,low-dose groups and the prozac 0.03g/kg group,each group of 12.For 7 days gavage,the suspension experiment was performed,and the accumulative immobile time of the last 4 min was recorded.The female Wistar rats were randomly divided into the normal control group,the pilocarpine control group,the high,medium and low-dose group,each group of 12.For 7 days gavage,in addition to the normal control group,the pilocarpine were injected under the skin of the rats(0.00035g/kg),and Hetian-gaoyuan reagent staining method was used to test the sweat of the rats in the palm.Results The ZYASP high-dose and diazepam can significantly increase the sleep rate of the hypnotic mice with subthreshold dose of napental(P<0.05,P<0.01);and the ZYASP low-dose and prozac can significantly shorten the accumulated inactivity time of the suspension test mice(P<0.05,P<0.01);at the same time,the high and medium-dose of ZYASP decreased the sweat number of the sweating model induced by pilocarpine in rats(P<0.05).Conclusion The ZYASP has the certain effect of hypnotic,anti-depression and antiperspirant.3 The intervention effect of ZYASP on the female climacteric syndromeObjective To observe the effect of ZYASP on menopausal symptoms and symptoms of kidney Yin deficiency.Methods 30 patients were screened with the patient’s various examination results and the diagnosis criteria of climacteric syndrome,and the treatment was given to the ZYASP,4 weeks was a course of treatment,with one course of treatment.To observe the changes of the Kupperman index score and the grading quantization score of the kidney Yin deficiency symptoms of the patients before and after the treatment and treatment began 3 months later.Results After treatment and treatment began 3 months later,the score of Kupperman index and the symptom of kidney Yin deficiency were significantly reduced(P<0.01),while the patient showed no obvious symptoms of discomfort and adverse of autonomic nervous function,and the symptoms of depression and anxiety,and has the effect of sedative-hypnotic and stop sweat;at the same time,the clinical efficacy is significant and the safety is high.The results of this study laid a foundation for further development of the new Chinese medicine of ZYASP,and its deep mechanism need to be further studied. |
