Objective:1.To understand the regularity of sex hormones changed gradually with aging which the people come from town resident of Changsha area, and the prevalence of male climacteric syndrome.2.To validated the hypothesis which HGF is material base of Yi-Guei-Tong-Yuan.3.To investigate the mechanism about Shen-Gan-Tong-Zhi intervene male climacteric syndrome.Methods:1.Using the questionnaire, cohere to biochemical indexes and the level of sex hormone, logistic regression analysis of related risk factors,protective factors.2.To constructed Leydig cells-liver nonpar-enchymal cells co-culture system, respectively in the drug containing plasma (Xiong can yi shen shu gan fang, Jin kui shen qi wan, Xiao yao san) diluted0times,4times,8times,16times after the intervention, which observed HGF, T content in the supernatant, HGF, c-Met, caspase-3index of cells.3. Replication of the male climacteric syndrome rat model, were randomly divided into model group, testosterone group, Jin kui group, Xiao yao group, Xiong can group, normal control group, after4weeks of continuous medication, to obsreved rat serum T, HGF, testis of T synthase (StAR, P450scc,3beta-HSD, HGF/c-Met), caspase-3index, the morpho-logical changes of testis tissue HE staining, the ultrastructure of interstitial cells of testis.Results:l.FT, Bio-T, TT in serum FTI, SHBG sex hormones changed gradually with aging which the people come from town resident of Changsha area, Male climacteric syndrome clinical total prevalence rate was10.85%, Age, body mass index, hypertension, smoking is the male climacteric syndrome risk syndrome factors, weekly regular exercise is a protective factor.2.HGF/c-Met signaling pathway may be involved in mediating T secreting, which Leydig cells-liver non parenchymal cells co-cultured. Xiong can yi shen Shu gan fang had the strongest effect.3.Male climacteric syndrome model of rat Leydig cell T synthesis capacity decreased, which may be related with the following mechanism:down regulated expression of T synthase (StAR, P450scc,3beta-HSD), down regulated expression of HGF/c-Met, up regulation of Caspase-3expression, Leydig cells apoptosis. Xiong can yi shen Shu gan fang had the strongest effect.Conclusions:With the increase of age, TT, FT, Bio-T levels decreased in which the people come from town resident of Changsha area, male climacteric syndrome with high incidence, regular exercise can effectively prevent. HGF/c-Met may be the key to the material basis of TCM Gan-Shen. Chinese medicine treatment of male climacteric syndrome, Gan shen tong zhi is better than Bu shen and Shu gan.
