ObjectiveThis study comprehensively collects Chinese medicine prescriptions for liver disease,building a database based on the Chinese medicine inheritance platform system and uses association rules and complex system entropy clustering methods to explore the prescriptions of Chinese medicine in the treatment of liver diseases.At the same time,the application of network pharmacology research methods,the study of mechanism of representative drug combinations aiming to clarify the law,provide some information data support and reference for the clinical rational and effective treatment of liver disease drugs and the development of new drug research.MethodA total 1181 prescriptions of treating liver diseases elected from TCM monographs were collected.A database was built relying on the "Chinese medicine inheritance auxiliary platform system",then we used the association rule apriori algorithm and complex system entropy clustering method to analyze data by using "data analysis system" and "prescription analysis system" from "statistical reporting system".Meanwhile,this analysis method was used to conduct an in-depth analysis of high-frequency drug prescriptions containing poria,salviae miltiorrhizae radix et rhizoma,atractylodis macrocephalae rhizome,artemisiae scopariae herba,and bupleuri radix,to figure out the association rules of each drug and drug combination in the prescription.Based on the 1181 TCM prescriptions for liver diseases,the chemical composition and target of" artemisiae scopariae herba,gardeniae fructus and rhei radix et rhizoma " drug pair were searched according to the TCMSP.Moreover,the protein interaction network of hepatitis C was established.Furthermore,establish the active component-predicted target network,GO functional enrichment analysis and enrichment analyses based on the KEGG pathway were constructed to study the drug treatment of hepatitis C.Result(1)Research of prescription using rule:a total of 1181 prescriptions for treating liver diseases in six TCMA monographs were collected.The frequency of use was ranked as follow:poria(579),salviae miltiorrhizae radix et rhizome(510),atractylodis macrocephalae rhizome(480),artemisiae scopariae herba(421),bupleuri radix(374).The top5 frequency of drug genus were:Asteraceae,Lamiaceae,Ranunculaceae,Apiaceae,Polyporaceae.Cooling drugs are the most used in prescriptions,followed by warm,neutral,cold and heat.The most frequent drug in prescription was pertained to liver channel,followed by the spleen channel and lung channel.High frequency drug combinations were:salviae miltiorr-hizae radix et rhizome and poria(259),atractylodis macrocephalae rhizome and poria(221),salviae miltiorrhizae radix et rhizome and paeoniae radix rubra(175),salvia miltiorrhizae radix et rhizome and curcumae radix(174),salviae miltiorrhizae radix et rhizome and artemisiae scopariae herba(169).The association rule with support degree 118,confidence>0.4 or higher includes:astragali radix->salvia miltiorrhizae radix(0.6525),atractylodis macrocephalae rhizome->poria(0.6424),astragali radix->poria(0.6144),codonopsis radix->poria(0.6142),paeoniae radix rzubra->salvia miltiorrhizae radix(0.5952)\The top five of the drug-based correlation analysis based on the improved mutual information method were:'puerariae lobatae radix,curcumae rhizome'(0.0119).?'sophorae flavescentis radix,xanthii fructus'(0.0116).?'rhei radix et rhizoma,margaritifera concha'(0.0114);approximately eighty drugs core combinations based on complex system entropy clustering;based on the new prescription analysis of unsupervised entropy hierarchical clustering,nearly 40new prescriptions were obtained.(2)Investigating the rule of medication prescription containing poria:the top three single drugs were poria,salvia miltiorrhizae radix,and atractylodis macrocephalae rhizome.the top three high-frequency drug combinations were:'salvia miltiorrhizae radix,poria','poria,atractylodis macrocephalae rhizome','bupleuri radix,poria',According to the support degree>20%,confidence>0.95association rules:salvia miltiorrhizae radix,astragali radix->poria','Salvia miltiorrhizae radix,bupleuri radix->poria','atractylodis macrocephalae rhizome,codonopsis radix->poria'etc.;based on the improved mutual information method,the leading three inter-drag association were:'dendrobii caulis,agrimoniae herba','rhei radix et rhizoma,polygoni cuspidati rhizome et radix','rhei radix et rhizoma,moutan cortex'.(3)Exploring the rule of medication prescription containing salvia miltiorrhizae radix:the top three single drugs are poria,salvia miltiorrhizae radix,and atractylodis macrocephalae rhizome.The top three high-frequency drug combinations were:'salvia miltiorrhizae radix,poria','salvia miltiorrhizae radix,paeoniae radix rubra','salvia miltiorrhizae radix,curcumae radix'.Association rules with support degree>20%,confidence=1 herba'.In the support degree>20%,confidence>0.98 association rules were:'imperatae rhizoma->artemisiae scopariae herba','angelicae sinensis radiX->artemisiae scopariae herba','salvia miltiorrhizae radix,bupleuri radix->artemisiae scopariae herba' etc.;based on the improved mutual information method,the leading three inter-drug association were:'sedi herba,phyllanthus urinaria','sedi herba,oxalis corniculata','paeoniae radix rubra,imperatae rhizoma'.(6)To explore the rule of medication prescription containing bupleuri radix:the top three single drugs were bupleuri radix,poria and salvia miltiorrhizae radix;the top three high-frequency drug combinations were:'salvia miltiorrhizae radix,bupleuri radix','poria,bupleuri radix','curcumae radix,bupleuri radix'.The association rules support degree>20%,confidence>0.98 shows:'codonopsis radix->bupleuri radix','glycyrrhizae radix et rhizoma->bupleuri radix','polygoni cuspidati rhizoma et radix->bupleuri radix'etc.;based on the improved mutual information method,the leading three inter-drug association were:'erodii herba geranii herba,glehniae radix','glehniae radix,perillae caulis','glehniae radix,gentianae macrophyllae radix'.(7)'artemisiae scopariae herba,gardeniae fructus,rhei radix et rhizoma'drugs pair are used to study the multi-component and multi-target network relationship of hepatitis C.It is preliminarily concluded that the active ingredient of this drug pair worked by 12 target including PIK3CG,CASP3,BCL2,CASP8 and MMP1;12signal pathways were obtained by KEGG pathway enrichment;GO functional enrichment analysis received4GO entries which were related to biological processes consist:negative regulation of apoptosis,positive regulation of cell proliferation,response to glucocorticoids and wound healingConclusionTCM treatment for liver diseases is ingenious,including activate blood to relieve pain,move qi to relieve,clear heat and resolve dampness etc.Network pharmacology method was used to study the multi-component and multi-target complex network relationship of'artemisiae scopariae herba,gardeniae fructus,rhei radix et rhizoma'drug pair.The results of the study preliminarily verified the mechanism of this drug pair on the treatment of hepatitis C,and provided information and foundation for further exploration.This study was hoping to provide a reference for TCM treatment of liver disease.
