| Objective: This animal experiment first by chronic heart failure model in rats and caused TCM model is a yang insufficiency syndrome,the rats were given saline,Yiqi Qiangyin Decoction and Benazepril hydrochloridehloride,carvedilol gavage,to explore when chronic heart failure,Yiqi Qiangxin Decoction in rats by acting on Ang II,NT-pro BNP and Caspase-3on the progression of chronic heart failure,changes of Ang II,NT-pro BNP and Caspase-3,to investigate the effects of Yiqi Qiangxin Decoction curative effect and mechanism of action in the treatment of chronic heart failure edema syndrome due to Yang deficiency in.Materials and methods: SPF class Wistar healthy male rats were provided by Liaoning Changsheng Biological Technology Co.,Ltd.,aged 10-12 weeks,weighing 80 185-220 g,divided into 20 sham operated group and 60 heart failure group.A model of chronic heart failure was induced by modified abdominal aortic coarctation in rats.After modeling,after 4weeks heart failure compensation period,the rat heart ultrasound was detected,and then the model rats were successfully injected with hydrocortisone acetate,which caused the deficiency of Yang in Chinese medicine.The heart failure rats were randomly divided into the model group,the Chinese medicine treatment group and the western medicine treatment group.The 3 groups and the sham operation group were divided into 4 groups.Chinese medicine Yiqi Qiangyin Decoction according to 22 G crude drug /(kg.d)dose of Western medicine benazepril hydrochloride by 10 mg /(kg.d)and dose of carvedilol by 6mg /(kg.d)dose gavage,sham operation group and model group received daily normal saline,continuous administration of 8 weeks.After the end of treatment,observe the general condition of rats,heart weight index of rats were measured,pathological changes of myocardium were observed with HE staining,ELISA detection of rat serum Ang and NT-pro BNP level II,detection of the expression of Caspase-3 in rat myocardium by immunohistochemistry,Effect of Yiqi Qiangxin Decoction on the possible mechanism of chronic heart failure and yang deficiency.Result:1.The general state of rats: Yiqi Qiangyin Decoction can improve the general state of chronic heart failure rats,improve the quality of life.2.rat heart weight index: Compared with sham operation group,model group HWI significantly increased(P <0.01).Compared with the model group,the western medicine group and the traditional Chinese medicine group were significantly lower(P <0.05,P <0.05).3.Changes of histomorphology in rats: Yiqi Qiangyin Decoction can significantly reverse the changes of heart and brain in rats with chronic heart failure,reduce the swelling of muscle fibers,disorder of myocardial cells,inflammatory cell infiltration and other myocardial pathological changes.4.The level of Ang ? in the serum of the rats was significantly higher than that of the sham group(P <0.01).Compared with the model group,the Ang ? of the western medicine group and the traditional Chinese medicine group were significantly decreased(P <0.05,P <0.05).5.NT-pro BNP levels in serum of rats: Comparison with sham group,model group NT-pro BNP were significantly higher(P <0.01).Compared with the model group,NT-pro BNP was significantly decreased in the western medicine group and the traditional Chinese medicine group(P <0.05,P <0.05).6.The expression of Caspase-3 in rats: compared with sham operation group,heart failure induced by myocardial overexpression of Caspase-3(P<0.01),compared with the model group,western medicine group,Chinese medicine group could decrease the Caspase-3expression(P<0.05,P<0.05).Conclusion:1 Yiqi Qiangxin decoction can improve heart function,reduce HWI,downregulation of Ang II,the level of serum NT-pro BNP.2.,Yiqi Qiangxin Decoction can reduce heart muscle cell apoptosis by reducing Caspase-3,so as to effectively improve heart failure.3.the mechanism of Yiqi Qiangxin Decoction in treating chronic heart failure and yang deficiency flood syndrome may be related to Ang II,NT-pro BNP and Caspase-3.It is proved that Yiqi Qiangxin Decoction plays an important role in the treatment of chronic heart failure. |
