Purpose:Through the intervention of detection of tear secretion,dry eye model rat after corneal fluorescein staining score,tear breakup time,corneal tissue pathology and serum IL-4,IFN-? content of gamma Zengyetang,dry eye to evaluate the effects of Zengyetang on dry eye model rats.To investigate the effect of decoction for increasing liquid on Th1/Th2 balance of dry eye model rats.Material and method:30 rats were fed for a period of up to 1 weeks.The rats were randomly divided into the control group(n=10)and the model group(n=20).The model group received a subcutaneous injection of 4 ? ˇml-1 scopolamine hydrobromide(8 o'clock,11o'clock,14 o'clock,17 o'clock,0.5ml each time,4 times a day)combined with corneal exposure preparation of dry eye model rats.The control group was injected with normal saline.Evaluation of dry eye model after continuous 28 d.The model rats were randomly divided into two groups: model control group(n=10)and decoction group(n=10).The blank control group did not do any treatment;The model control group was given the same amount of physiological saline every day,and the decoction group was treated with intragastric administration of 8.125g/kg(1.625g/ml,1ml)every day.Until the end of 56 d gavage.The amount of tear secretion,tear film break-up time and corneal fluorescein staining were measured in each group.Hematoxylin eosin(HE)staining was used to observe the pathological changes of corneal tissue in rats,and the levels of IL-4 and IFN-? in serum were detected by enzyme linked immunosorbent assay(ELISA).Results:1.The twenty-eighth day after injection of scopolamine,using tear detection filter strips on tear secretion test two groups of rats,the results showed that length of wetting tear model rats detection filter strips significantly less than the blank control group(p<0.01);application of fluorescein sodium eye test and determination of tear film slit lamp rats rupture time.The breakup time of tear film in model group was significantly less than that in blank control group(p<0.01).The fluorescein sodium staining score of each group was measured by fluorescein sodium ophthalmic test paper and slit lamp.The corneal fluorescein sodium staining score of model group was significantly higher than that of control group(p<0.01).2.At 2 hours after the end of the last intragastric administration,the tear secretion of rats in each group was measured.The measurement results are as follows:compared with the blank control group,model control group and treatment group of Zengyetang tear moist length detection filter strips significantly reduced,there was a statistically significant difference(p<0.01);compared with model control group,Zengye Decoction group tear moist filter paper test length increased significantly and there was a statistically significant difference(p<0.01).3.At 2 hours after the end of the last intragastric administration,the break-up time of the rats in each group was measured.The measurement results are as follows:compared with the blank control group,model control group and treatment group Zengyetang break-up time was shortened obviously,there was a statistically significant difference(p<0.01);compared with model control group,Zengye Decoction group break-up time significantly increased,there was a statistically significant difference(p<0.01).4.At 2 hours after the end of the last intragastric administration,the corneal sodium fluorescein score was applied to each group.The measurement results are as follows:compared with the blank control group,model control group and treatment group of Zengyetang corneal fluorescein staining score was significantly increased,there was a statistically significant difference(p<0.01);compared with the model control group,Zengye Decoction group corneal fluorescein staining score was significantly decreased,there was significant significant(p<0.01).5.The pathological observation of the three groups of rats were as follows:the blank control group:the level of cornea was clear.The morphology of epithelial cell layer was normal,and the cell morphology was normal and orderly.Model control group:irregular proliferation of epithelial cells,abnormal arrangement of cell morphology disorder,corneal surface keratosis.Layered fuzzy,flat cells appear to fall off,visible wing cells exposed.A large number of inflammatory cell infiltration,edema thickening.Zengyetang treatment group:only mild hyperplasia of the corneal surface.The morphology of the cells was good,and the arrangement of the cells was good.The basement edema disappeared and the inflammatory infiltration decreased.6.Analysis of serum IL-4,IFN-? content,the results are as follows:IL-4:compared with the blank control group,model control group and Zengyetang treatment serum in IL-4 group was significantly reduced,there was a statistically significant difference(p<0.01);compared with the model control group,Zengye Decoction in the treatment of serum the content of IL-4 was significantly increased,there were significant statistical difference(p<0.05).IFN-?:compared with the blank control group,model control group and Zengyetang treatment group serum IFN-? content increased,there was a statistically significant difference(p<0.05);compared with the model control group,Zengye decoction treatment group serum IFN-? content decreased significantly,there was significant difference(p<0.01).Conclusion:1.Dry eye model by subcutaneous injection of scopolamine can replicate the dry eye model,the model making method of dry eye model with strong stability(for long-term experiments),molded short time,simple operation,high achievement ratio,the mortality rate is low,the advantages of wide application range.2.The curative effect of decoction for increasing fluid was observed:after application of the decoction,the amount of tear secretion increased,tear film break-up time(BUT)prolonged,corneal fluorescein staining score decreased,corneal pathology also became normal.The effect of liquid soup.3.The mechanism of decoction for treating dry eye:in the process of dry eye,the content of IL-4 in serum and the content of IFN-? increased,and the Th1/Th2 balance was destroyed.Zengyetang can increase the content of IL-4 in the serum of model mice and reduce the content of IFN-?,which makes the balance of Th1/Th2 to normal,which may be one of the reasons for the treatment of dry eye.
