Objective: Shows Jin kiu type STC rat model was established,the detection of constipation AQP3 in the colon of rats,AQP9 and 5-HT,the expression of NOS in serum,to study the effect of increasing liquid soup play aperient effect mechanism.Methods: 36 SD rats were randomly divided into 6 groups,respectively is: the blank group(don't die,don't treatment);Model group B(only build mode: limit water + compound diphenoxylate,no treatment);Western medicine group(model C + HCL will benefit treatment group);Low dose group D(model + add liquid soup lower dose treatment groups);Traditional Chinese medicine(TCM)measurement in group E(model + add liquid soup dose in the treatment group);Chinese medicine high dose group(model F + add liquid soup high dose treatment group);Using compound diphenoxylate(10 mg/kg)for B,C,D,E,F group rats lavage for 7 days,constipation model is set up,executed A,B group of rats at the end of the more general situation,the waste water content,carbon propulsion rate to verify the model.later,hydrochloric acid will benefit(50mg/kg)to lavage 6 days,group C will increase enrichment configured to a certain proportion of liquid liquid soup,d,E,F group were given 0.5 ml + 2.5 ml of liquids ultrapure water,ultrapure water 1 ml + 2 ml of liquids,2 ml + 1 ml of liquids ultrapure water(equal to 5 times dosage of adult day,10 times,20 times)to rats lavage,Ed)6 days.All rats to be put to death before the ink suspension lavage,determination of ink advance rate.Using immune histochemical method,ELISA to detect the rat proximal colon tissue of AQP3,AQP9 expression and activity of NOS,5-HT level.Results:Intervention in the treatment of Jin kiu type STC after different concentration increases the liquid soup rats,colon tissue of immunohistochemical results show:AQP3 in group B MD value is greater than in group A,group C,group D,E,F group,the MD value(p < 0.05),the difference was statistically significant;Group C compared with group D(p > 0.05),no statistical significance;Group C compared with group F MD value(p < 0.05),the difference was statistically significant.AQP9 in group B MD value is less than group A,group C,group D,E,F group,the MD value(p < 0.05),the difference was statistically significant;Value AQP9 MD in group A were big group C,group D,E,F group,the MD value(p < 0.05),the difference was statistically significant.ELISA to detect serum concentration of 5-HT,group B is less than the other five groups(p < 0.05),the difference was statistically significant.F less than C-E group(p < 0.05),the difference was statistically significant;Group A compared with group F(p > 0.05),there was no statistically significant difference.And the concentration of NOS,group B is greater than the other 5 groups(p < 0.05),the difference was statistically significant;Group A is less than C-F group(p < 0.05),the difference was statistically significant;Group C compared with group D(p > 0.05),there was no statistically significant difference.Conclusion:(1)Increased colon epithelium AQP3 expression,expression AQP9 is reduced,serum 5-HT reduce and NOS content increases,leading to excessive absorption of moisture,colon intestinal juice secretion decreased,may be one of the pathogenesis of on STC.(2)Increased by cutting liquid soup AQP3 expression,raise AQP9 expression,raise serum 5-HT and reduce NOS level,could be a play to push the boat "effect of" that can be one of the mechanism of action of treatment on STC.
