The Effect Of Orexin-A Combined With Anti-emetics On Cisplatin-induced Rat Ecstasy

Objective: To observe the clinical effect of improved stop vomiting drug ondansetron,aprepitant or pemirolast combined with intracerebroventricular injection of Orexin-A on cisplatin induced pica in rats with anorexia,and further explore the mechanism of pemirolast Orexin-A combined with anti vomiting.Methods: Preparation of cisplatin induced pica model rats,intraperitoneal injection of different dose of ondansetron,aprepitant or pemirolast,and lateral ventricle puncture and injection of Orexin-A in rats of kaolin intake and food intake changes.EIA method was used to measure the content of substance P in cerebrospinal fluid.Results : Intraperitoneal injection of high dose cisplatin in rats,kaolin intake significantly increased,decreased food intake,body weight decreased significantly,and showed a significant dose-dependent manner(P < 0.05~0.01);ondansetron early inhibition of cisplatin in rats of kaolin intake(P < 0.05),combination of intracerebroventricular injection of Orexin-A can be extended.The effect can be increased,and the late food intake of rats(P < 0.05);aprepitant can inhibit cisplatin in rat advanced kaolin intake,and a significant dose dependent relationship(P < 0.05~0.01),fourth days after the intracerebroventricular injection of Orexin-A combined with cisplatin in rats,kaolin intake was significantly reduced(P < 0.05),and food intake increased significantly(P < 0.05);application of pemirolast,intake of cisplatin can significantly inhibit the rat advanced kaolin,and showed a significant dose-dependent manner(P < 0.05~0.01),high dose of pemirolast application,But cisplatin rat advanced food intake increased significantly(P < 0.05),fourth,5 days after intracerebroventricular injection of Orexin-A combined with cisplatin in rats,kaolin intake significantly reduced food intake,first,4,5 days increased significantly(P < 0.05);cisplatin in cerebrospinal fluid of rats significantly increased the content of substance P(P < 0.05),pemirolast combined with intracerebroventricular injection of Orexin-A,cisplatin in rat cerebrospinal fluid substance P content decreased significantly(P < 0.05),suggest that the effect may be related to the synthesis or release of substance P.Conclusions: The ondansetron,aprepitant or pemirolast in combination with Orexin-A,can extend the antiemetic drug effect,and can improve the cisplatin induced anorexia,the effect may be related to substance P signaling pathway..
