Object: To establish cisplatin-induced pica in rat, as a chemotherapy-induced neausea and vomiting(CINV) model for the first time in mainland China, to observe the preventing and treating effects of the aqueous extract of Xiao-Ban-Xia-Tang (Little Pinella Rhizome Decoction,小半夏汤). Methods: First, Wistar rats were housed individually in cages with plastic–mesh cap to supply kaolin pellets, food and water were also available and libitum. 72h later, rats were injected intraperitoneally with cisplatin to induce pica. Subsequently, the consumption of kaolin,food,water and body weight were recorded. 5-HT's expression of rats'ileum was observed by Envision immunohistochemistry. Pica behaviour was considered as the degree of nausea and vomiting. Results: Different dose of cisplatin was injected intraperitoneally to induce pica. In contast to the control group, except for 3mg/kg, 6mg/kg,7.5mg/kg and 9mg/kg group of cisplatin injected intraperitoneally could cause 24h kaolin consumption. During 72-96h, 9mg/kg group animals died, which indicted that 9mg/kg of cisplatin is too high for rats to endure. The consumption of kaolin,food,water and body weight were also reduced by cisplatin. 7mg/kg is considered as the appropriate dose to induce pica. The aqueous extract of Xiao-Ban-Xia-Tang could reduce kaolin consumption, modify food and water intaken, reduce the 5-HT's expression of rats'ileum and improve the regulation of gastric emptying. Conclusions: The present studies indicated that pica can be induced by cisplatin in rat,and the aqueous extract of Xiao-Ban-Xia-Tang can inhibit chemotherapy-induced nausea and vomiting (CINV),improve rats'status.
