Objective:To investigate the prevention and treatment effect and 5-HT mechanism of Xiao-Ban-Xia-Tang (Little Pinella Rhizome Decoction) in chemotherapy-induced rat pica model. Methods:Emetic stimuli induced kaoli ingestion behavior (pica behavior) in rats (species that do not have the emetic reflex) is analogous to gastrointestinal discomfort, such as nausea and emesis. The pica behavior is served as index of nausea and vomiting. Consumption of kaolin in rats was used as an index of nausea and vomiting. The antiemetic effect and 5-HT mechanism of Xiao-Ban-Xia-Tang were observed in the methods of rat pica models induced by cisplation. The gray value of 5-HT,5-HT3A receptor positive cells of rat ileum and brain was detected in the immunohistochemistry, and the levels of 5-HT,5-HIAA in serum and brain of rats were determinated in reversed-phase high performance liquid chromatography. Results:The aqueous and ethanol extraction of Xiao-Ban-Xia-Tang significantly inhibited consunmption of kaolin in rats, and the ethanol extraction demonstrated a better curative effect; The aqueous and ethanol extraction of Xiao-Ban-Xia-Tang reduced the expression of 5-HT,5-HT3A receptor positive cells in rat ileum and brain, and depressed the levels of 5-HT,5-HIAA in rat serum and brain. Conclusion:Xiao-Ban-Xia-Tang can significantly prevent and treat the chemotherapy-induced nausea and vomiting, and its mechanism may be related to inhibiting 5-HT synthesis,release, and down 5-HT3A receptor expression.
