Purpose:By intranasal inoculation,the establishment of IV(FM1 strain)in Wistar rat model of acute lung injury in young infected with lung tissue pathology;evaluation score of lung tissue injury,the levels of p-ERK,t-ERK,NF-kappa B p65 expression in lung tissue was measured by immunohistochemistry and Western blotting,in-depth study and study of IV infection in rats after lung injury and external application of Chinese medicine treatment of influenza virus pneumonia effect mechanism,to provide a theoretical basis for the clinical application and therapeutic methods of TCM and Chinese medicine development.Material and method:1.The influenza A virus FM1 strain inoculated LD50 concentration method,establish the model of acute lung injury in young Wistar rats infected with influenza virus,observe the general condition of rats by day(diet and water intake,hair,activity,mental state,body weight,respiratory rate),lung tissue gross appearance observation,study pathological observation of lung tissue under light microscope,the lung tissue pathological score multiple angle IV virus pneumonia identification model.2.With high,middle and low dose of Tongluo Qingfei Fuxiong plaster on the back in juvenile rat as the route of administration,intervention on pneumonia rat model induced by IV,we observed Wistar general condition,lung tissue biopsy,and the lung pathological score,lung index of high dose group,middle dose group,low dose group of Tongluo Qingfei Fuxiong ointment,a comparative study of influenza virus pneumonia treatment.3.The phosphorylation of ERK growth rate and lung tissue in p-ERK,t-ERK,NF-K B 3 p65 protein expressed by immunohistochemical method and Western blot method to detect Qingfei Tongluo Fuxiong pluvis treatment group Wistar in juvenile rat lung tissue,to investigate the acute pneumonia Tongluo Qingfei Fuxiong ointment in the treatment of IV induced treatment relationship therapy mechanism and dose.Results:1.IV Wistar infection in juvenile rat model identification results: the model control group made in poor mental state,weight loss,weight growth is slow,eating and drinking are significantly reduced,weight loss,body hair stands without luster,curled up,back,slow reaction,slow action,activity decreased,respiratory rate increased soon,the lung tissue appearance color red,hyperemia and edema,compared with normal control group,the lung weight imbalance,observed the injury of alveolar epithelial cell shedding pathological staining,pulmonary interstitial and perivascular edema thickening,alveolar exudation of inflammatory cells.2.Comparison of body weight and lung index in rats: the rats in the model group weight is the slowest,Qingfei Tongluo Fuxiong ointment in the treatment of group weight increased in high dose group increased most rapidly,there are obvious differences between the normal group(p<0.05);compared with the model group,the other rats lung index were significantly decreased(p<0.05);in low dose group,high dose group decreased lung index compared with no obvious.3.Compared with the normal control group,model group and treatment group in the injury area,trachea,alveolar capillary inflammation around the ratio of the amount of inflammatory score had significant difference(p<0.05);high dose group pathological scores were significantly less than the low dose group,there are significant differences with the model group(p<0.05).There was a significant difference between the two groups(p<0.05).There was no difference between the low-dose group and the model group(p>0.05).Promptqingfei Tongluo Fuxiong plaster can reduce the lung injury induced by IV,and the dose level of treatment effect,with the most obvious therapeutic effect of high dose group.4.Compared with the normal control group,lung tissue of model group and treatment group in p-ERK,NF-kappa B p65 expression had significant difference(p<0.05);the expression level of p-ERK protein in lung tissue,Tongluo Qingfei Fuxiong ointment in the treatment group and model group were significantly different(p<0.05),the high dose group than in low dose group;expression the level of NF-in B kappa p65 protein in lung tissue,high dose group and medium dose group was significantly lower than that of model group(p<0.05),there was significant difference with the low dose group(p<0.05);low dose group in the lung tissue of NF-kappa B p65 expression had no significant difference with the model group(p>0.05).5.The lung tissue of model rats in the phosphorylation of ERK protein increased significantly compared with the normal group(p<0.05);high,medium and low dose group of ERK protein phosphorylation of Qingfei Fuxiong pluvis Tongluo was significantly inhibited,and there was significant difference compared with model group(p<0.05),middle dose group and the high-dose group of ERK protein phosphorylation level with the high dose group significantly(p<0.05),and has the obvious difference with the low dose group(p<0.05).That activation of Fuxiong pluvis Tongluo Qingfei could inhibit ERK protein in rat lung tissue,thereby reducing inflammation in lung tissue.6.By the method of single factor analysis,the results showed that the model group of p-ERK,NF-kappa B p65 protein expression level is the highest,there are significant differences with other groups(p<0.05),the expression of p-ERK in the high dose group,NFkappa B p65 protein levels were significantly lower,obviously different with medium and low dose group(p< 0.05).The expression of NF-kappa B protein in the middle dose group was significantly different from that in the low dose group(p65).Conclusion:1.by intranasal inoculation of influenza virus strain FM1 in young rats,the general state of rats,observe the histopathologic observation,lung index and lung tissue pathological score,successfully identified pneumonia rat model induced by IV.2.Tongluo Qingfei Fuxiong ointment can significantly reduce the lung tissue pathological score and lung index IV pneumonia,improve the general state of young rats,the pathological changes in lung tissue inflammation,reduce IV infection caused by inflammation of the lung tissue pathological injury3.Fuxiong ointment can Tongluo Qingfei by inhibiting ERK/ activation of NF-B signaling pathway,reduce viral replication,reduce inflammation of lung tissue,the antiviral effects of.The expression of ERK and NF-kappa B protein was lowest in the high and middle dose groups in the course of viral pneumonia(p65).
