Purpose: To establish a Mycoplasma pneumoniae pneumonia(MPP)model in BALB/c mice infected with Mycoplasma pneumoniae(MP);application of Qingfei Ointment(Superfine granule,Ordinary granule)in the dorsal shu area of mice was used to study the regulation mechanism of the TLR2/NF-?B signaling pathway of the MPP mice induced by MP and to explore its safety.Material and method: Screening SPF BALB/c mice 96,randomly divided into normal group,model group,Qingfei Ointment ?Group,Qingfei Ointment ? Group,each group of 24,in addition to the normal group,all the remaining groups were using mycoplasma bacterial liquid drop mice nasal reach lower respiratory tract infection mechanism to establish MPP mice model.After the mould was successful,the mice in the normal group and the model group were given the corn Flour on the back of the dorsal Shu Acupoint,The Qingfei Ointment ?Group and Qingfei Ointment ? Group were given the superfine granule Qingfei Ointment,Ordinary granule Qingfei Ointment external to the back of the dorsal Shu Acupoint,apply area 3x2cm2,paste once a day,each 30 min,for 14 consecutive days.After the modelling 3,7,10,14 days the group of mice were executed in batches,the pathological changes and pathological changes of the lungs were observed,the lung index was measured by weighing method,the expression of TLR2 in mice lungs was detected by q PCR method,and the serum My D88 in mice was detected by ELISA.The expression of NF-?B in mice lung tissue was detected by IL-6 and TNF-? levels and Western blot method.Results: 1.After MP infection,the pathological sections of lung tissue in normal group were normal.Inflammatory cell infiltration in mice lung tissue in model group alveolar structure destruction and other changes,the 7th day is the most obvious,14 days when inflammation gradually reduced;after treatment,Qingfei Ointment ?Group,? Group of mice were in the 3rd and 7 days after the model was significantly reduced,The difference was significant(p<0.05),compared with Qingfei Ointment ? Group,the pathological score of Qingfei Ointment ? group decreased significantly in the 10 th day,The difference was statistically significant(p<0.05),and the 3rd and 14 th days were compared,the pathological scores of each group were significantly different(p<0.05),compared with the control group,the treatment group used two different dosage forms of Qingfei Ointment applied to the dorsal of the mice in the dorsal-Shu Acupoint area,the local skin of the mouse to observe without redness,blood infiltration,under the microscope observation,The epidermis structure of mice in control group and treatment group was normal.2.After MP infection,the Lung index of model group mice and normal group increased markedly,in the 7th day peak,the difference was statistically significant(p<0.05),the lung index decreased after treatment,Qingfei Ointment ?Group,Qingfei Ointment ? Group of mice after treatment 3,7 days compared to the model group have significant difference(p<0.05);compared with Qingfei Ointment ?,the lung index of Qingfei Ointment ? group decreased significantly in the 7th day,and was statistically significant(p<0.05).On the 3rd and 14 th days,there was no significant difference in Lung index(p>0.05)between the groups.3.After MP infection,the expression of TLR2 in the model group increased,and the peak was the 7th day after the mould was made.All the time points were statistically significant compared with the normal group(p<0.05),after treatment,the expression of TLR2 decreased,the 3rd day two treatment group effect is not obvious(p>0.05),7th day Qingfei Ointment ? group began to effect,TLR2 expression decreased(p<0.05),and Qingfei Ointment ? Group has not yet been effective(p>0.05),10 th day Qingfei Ointment ? Group T LR2 expression also decreased,but not as Qingfei Ointment? Group effect significantly,the difference is obvious(p<0.05),the 14 th day two treatment group can significantly reduce TLR2 expression,there is no significant difference between the two groups(p>0.05).4.After MP infection,the content of My D88 in the model group increased,the 10 th day after the mould was peaked,and then gradually decreased.All the time points were statistically significant compared with the normal group(p<0.05),after treatment,the My D88 content decreased,the 3rd day two treatment group effect is not obvious(p>0.05),the 7th day Qingfei Ointment ? Group starts to effect,the My D88 content has reduced(p<0.05),but the Qingfei Ointment ? Group has not been effective(p>0.05),the 10 th day Qingfei Ointment ? Group My D88 content also decreased,but not as Qingfei Ointment ? Group effect significantly,the difference is obvious(p<0.05),the 14 th day two treatment group can significantly reduce My D88 expression,there is no significant difference between the two groups(p>0.05).5.After MP infection,the expression of NF-?B protein in the model group increased,the 3rd day after the mould was peaked,and then gradually decreased,Each time point was statistically significant compared with the normal group(p<0.05),after treatment,the expression of NF-?B decreased,the 3rd day two treatment group effect is not obvious(p>0.05),the 7th day Qingfei Ointment ? Group is effective,NF-?B expression has decreased(p<0.05),and Qingfei ointment ? Group has not been effective(p>0.05),10 th day Qingfei Ointment ? Group NF-?B also decreased,but not as Qingfei Ointment ? Group effect significantly,the difference is obvious(p<0.05),the 14 th day both treatment group can significantly reduce NF-?B expression,there is no significant difference between the two groups(p>0.05).6.After MP infection,the content of IL-6 and TNF-? in the model group was higher than that in the 7th day,and then gradually decreased,but still higher than normal group,each time point and normal group were significantly different(p<0.05),the IL-6 and TNF-? content of the two treatment groups decreased,On the 3rd day,the effect of the two treatment group was not obvious(p>0.05),the 7th day Qingfei Ointment ? group start to effect,IL-6,TNF-? content all drops(p<0.05),but the Qingfei Ointment? Group has not been effective(p>0.05),10 th day Qingfei Ointment? group IL-6,the TNF-? content also reduces,But it is inferior to Qingfei Ointment ? group Curative effect is remarkable,the difference is obvious(p<0.05),the 14 th day two treatment group can obviously reduce IL-6,TNF-? content,the comparison between two groups is not obvious difference(p>0.05).Conclusion: 1.Compared with the ordinary granules,the Superfine granule Qingfei Ointmentcan reduce the inflammation of the lung tissue of MPP mice,and its effect is more obvious and safe and effective in the later stage of MP infection 2.The expression of TLR2 in MPP mice lung tissue was basically consistent with that of My D88 and NF-?B,and it was speculated that MP could be involved in inflammatory reaction by relying on TLR2/NF-?Bsignaling pathway.Qingfei Ointment can reduce the expression of TLR2 and NF-?B protein in the lung tissue of MPP mice,It can also decrease the content of My D88,IL-6 and TNF-? in serum.3.The mechanism of treat MP infection of Qingfei Ointment may be related to reducing the release of proinflammatory factors and regulating the conduction of TLR2/NF-?B signaling pathway.
