BackgroundPeople are enjoying efficient and convenient life style from the progress,development and continuous changes of society,but also they are bearing more and more pressure.Especially for people who are living in cities,they are facing pressures from work,study and carrier,as well as fiercer competition.At this point,people tend to be more prone to acute and chronic stress.Moderate stress can improve the body's ability to adapt and maintain the motivation,but excessive stress lasted for a long time,and it will become the burden of adaptation,which will lead to anxiety if lasting for long time.Studies have shown that long-term stress could lead to anxiety.80%of the diseases are closely related with stress currently.In the stress state,the disorder of the neuroendocrine system leads to abnormal secretion,synthesis and transfusion of the relevant inflammatory cytokines.Heat shock protein 70(HSP70),as an important protective protein,is involved in the regulation of inflammatory cytokines.The clinical anxiolytic drugs have certain side effects.Researchers gradually focus on traditional Chinese medicine therapyObjective.This study aims to investigate the effect of Xiaoyao Powder on gastric mucosa HSP70 and related inflammatory cytokines in rats with chronic restraint stress anxiety model,and to explore the mechanism of Xiaoyao Powder in protecting gastric mucosal injury caused by emotional internal damage,as well as to provide experimental basis for the clinical application of Xiaoyao Powder in the treatment of gastric mucosal injury caused by emotional discomfort.Methods48 healthy SD rats weighing from 220-250g were randomly divided into normal group,model group,Xiaoyao Powder group and Rolipram group,with 12 in each group.Adaptive feed was given to them for 10 days.The experiment group used chronic restraint stress to induce anxiety rat model for 3 hours a day and continuously 14 days.Xiaoyao Powder group was received intragastric administration by Xiaoyao Powder suspension liquid,while normal group and model group were received intragastric administration by deionized water and Rolipram injection drugs respectively.The rats were subjected to body weight measurements at days 0,7,and 14.The behavioral tests were performed with the elevated cross maze.A novel inhibition experiment was performed on days 0 and 14 of the experiment.Rats' general situations of mental state,body posture,fur color,activity,the degree of response to the restrain,eye mucosa color,auricle color and feces and others were observed every day.The expression of heat shock protein 70 was detected by immunohistochemistry.The levels of IL-6,IL-8 and other related inflammatory cytokines in serum were measured by ELISA.Results1.General situation.At the end of two weeks' modeling,the rats in normal group,Xiaoyao Powder group and Rolipram group had more swiftly motion,softer squeak,and the times of screaming,fighting and struggling were significantly less compared with that in model group.Rats had stable mental condition and neater hair compared with model group,but more messy than normal group.Feces were in moderate dry and wet,and in ball shape.However,rats in model group were in low emotion,with messy hair and dark color,crouching in the corner with vigilant state.Resistance,screaming and struggling when in resistance were slightly reduced than a week ago.2.For the weight changes,there was no significant difference in weight 1 day before modeling and 7 days after modeling.However,weight of rats in the other three groups was lighter than normal group on the 14th day,and the difference was statistically significant(P<0.05).Compared with normal group,there was no significant difference in weight of rats in Xiaoyao Powder group and Rolipram group.3.In the experiment of intake resistance in new environment,there was statistical difference among groups at the baseline.And on the 14th day after modeling,the time in model group was significantly longer than Xiaoyao Powder group.4.For the elevated cross maze,at the end of two weeks' modeling,the number of open arms entering was reduced and the residence time was shortened in model group when compared with normal group,while the number was increased and time was prolonged in Xiaoyao Powder group and Rolipram group compared with model group without statistical difference.5.In immunohistochemistry experiment,the expression of HSP70 in the gastric mucosa of the model group was significantly lower than that in the normal group,which was statistically significant(P<0.05).Compared with model group,the positive expression in Xiaoyao Powder group was significantly increased,and it was statistically significant(P<0.01).While there was no statistically significant difference in Xiaoyao Powder group and Rolipram group compared with normal group,the expression of Xiaoyao Powder group was slightly higher than that in Rolipram group.6.In ELISA experiment,the levels of IL-1?,IL-6,IL-8 and TNF-a in the serum of model group were significantly higher than those of the other three groups.Compared with normal group,there was no statistically significant difference between Xiaoyao Powder group and Rolipram group.Conclusion14 days chronic restraint stress could lead to anxiety in rats.It suggesting that Xiaoyao Powder could promote gastric mucosa to produce a large number of HSP70 to inhibit the release of inflammatory cytokines,thus play a role in the protection of gastric mucosa,which provide experimental basis for the clinical application of Xiaoyao Powder in the treatment of gastric mucosal injury caused by emotional discomfort.
