Diabetes mellitus is a diseases that associated with multiple metabolic disorders,t he main feature of which is chronic hyperglycemia caused by insufficient insulin secre tion and action.The pathogenesis of Diabetes include both genetic and environmental factors.Now DM has became a serious worldwide health problem due to its high inci dence and mortality rate,and also a heavy burden of medical expenses to many count ries.More and more researchers concentrate on treatment and mechanism of DM.Mor inga oleifera leaves,indigenous to India,is a traditional herb that has various therape utic effects such as:hypoglycemic,hypotensive,anti-inflammatory and antibacterial.No w many researches have proved its hypoglycemic effect,but the mechanism is still un revealed.No report suggests if Moringa oleifera leaves lower blood glucose level by affect the gut microbiota.This study will observe the change in gut microbiota of ZD F rats to explore the hypoglycemic mechanism of Ethanol extracts of Moringa oleifera Leaves(EoM).ObjectiveUsing spontaneous diabetes mellitus ZDF(fa/fa)rats to observe the effect of Eo M on blood glucose and gut microbiota,to explore the hypoglycemic mechanism of E oM.Method10 male ZDF(fa/+)rats were used as normal group,40 male ZDF(fa/fa)model ed rats were randomly divided into four groups:control group,metformin group,EoM low dose group and EoM high dose group,10 for each.Intragastric administration fo r 8 weeks,measured blood glucose every week and body weight every two weeks.O GTT and ITT were tested in the 5th week and the 6th week.GLU,TG,TC,HDL and LDL of serum were measured after sacrificed.Fresh fecal samples were collected in the 2nd week(control group only)and the 8th week(all groups)for 16S rDNA high t hroughput sequencing to analyze the changes in gut microbiota.Results8 weeks after administration,the body weight gain of 4 ZDF diabetic rats groups were significantly lower than the normal group(p<0.05),and the body weight gain o f EoMh group was significantly lower than that of control group(p<0.05);EoMl group,EoMh group and Met group had significant effect on lower serum GLU level(p<0.05,p<0.01 and p<0.01),EoMh can also lower the serum LDL level and TG level.16S rDNA high-throughput sequencing showed that the diversity of gut microbiota in cont rol group was significantly decreased(p<0.01);EoMh significantly increased the gut mi crobiota diversity of ZDF diabetic rats(p<0.05).Both EoMI and EoMh can significantl y reduce the abundance of Bacteroidetes,Prevotellaceae and Prevotella(p<0.05),and si gnificantly increased the abundance of Firmicutes,Lactobacillaceae and Lactobacillus(p<0.05).EoMh also significantly increased the abundance of Ruminococcaceae and Lac hnospiraceae(p<0.01 ? p<0.05).ConclusionsEoM can lower the blood glucose level of ZDF diabetic rats,EoMh can also dec rease the blood lipid level,EoM has the effect of treating diabetes;EoM can increase the diversity of intestinal microflora in ZDF diabetic rats and improve the distributio n of gut microbiota,and high dose has a more significant effect;the effect of regulati ng the gut microbiota may be the hypoglycemic mechanism of EoM regulating the gu t microbiota.
